Human
adipose
depots
are
functionally
distinct.
Yet,
recent
single-nucleus
RNA
sequencing
(snRNA-seq)
analyses
largely
uncovered
overlapping
or
similar
cell-type
landscapes.
We
hypothesized
that
adipocyte
subtypes,
differentiation
trajectories
and/or
intercellular
communication
patterns
could
illuminate
this
depot
similarity–difference
gap.
For
this,
we
performed
snRNA-seq
of
human
subcutaneous
visceral
tissues
(five
ten
samples,
respectively).
Of
27,665
nuclei
in
both
depots,
most
were
'classical',
namely
enriched
lipid
metabolism
pathways.
However,
also
observed
'nonclassical'
immune-related,
extracellular
matrix
deposition
(fibrosis),
vascularization
angiogenesis
ribosomal
and
mitochondrial
processes.
Pseudo-temporal
analysis
showed
a
developmental
trajectory
from
progenitor
cells
to
classical
adipocytes
via
nonclassical
adipocytes,
suggesting
the
state
stems
loss,
rather
than
gain,
specialized
functions.
Last,
routes
consistent
with
different
inflammatory
tone
two
depots.
Jointly,
these
findings
provide
high-resolution
view
into
contribution
cellular
composition,
fat
differences.
Single-nucleus

Abstract
Over
the
past
few
decades,
RNA
sequencing
has
significantly
progressed,
becoming
a
paramount
approach
for
transcriptome
profiling.
The
revolution
from
bulk
to
single-molecular,
single-cell
and
spatial
approaches
enabled
increasingly
accurate,
individual
cell
resolution
incorporated
with
information.
Cancer,
major
malignant
heterogeneous
lethal
disease,
remains
an
enormous
challenge
in
medical
research
clinical
treatment.
As
vital
tool,
been
utilized
many
aspects
of
cancer
therapy,
including
biomarker
discovery
characterization
heterogeneity
evolution,
drug
resistance,
immune
microenvironment
immunotherapy,
neoantigens
so
on.
In
this
review,
latest
studies
on
technology
their
applications
are
summarized,
future
challenges
opportunities
discussed.

Understanding
gene
function
and
regulation
in
homeostasis
disease
requires
knowledge
of
the
cellular
tissue
contexts
which
genes
are
expressed.
Here,
we
applied
four
single-nucleus
RNA
sequencing
methods
to
eight
diverse,
archived,
frozen
types
from
16
donors
25
samples,
generating
a
cross-tissue
atlas
209,126
nuclei
profiles,
integrated
across
tissues,
donors,
laboratory
with
conditional
variational
autoencoder.
Using
resulting
atlas,
highlight
shared
tissue-specific
features
tissue-resident
cell
populations;
identify
that
might
contribute
neuromuscular,
metabolic,
immune
components
monogenic
diseases
biological
processes
involved
their
pathology;
determine
modules
underlie
mechanisms
for
complex
traits
analyzed
by
genome-wide
association
studies.
