The novel coronavirus disease (COVID-19) outbreak that emerged at the end of 2019 has now swept world for more than 2 years, causing immeasurable damage to lives and economies world. It drawn so much attention discovering how severe acute respiratory syndrome (SARS-CoV-2) originated entered human body. current argument revolves around two contradictory theories: a scenario laboratory spillover events contact with zoonotic diseases. Here, we reviewed transmission, pathogenesis, possible hosts, as well genome protein structure SARS-CoV-2, which play key roles in COVID-19 pandemic. We believe was originally transmitted by animals rather leak. However, there still needs investigations determine source Understanding is vital developing global strategies mitigating future outbreaks.

The number of genetic variations in the SARS-CoV-2 genome has been increasing primarily due to continuous viral mutations. Here, we report that human APOBEC3A (A3A) cytidine deaminase plays a critical role induction C-to-U substitutions genome. Bioinformatic analysis chronological changes sequence database indicated largest UC-to-UU mutation signature, consistent with APOBEC-recognized nucleotide motifs, was predominant single-stranded RNA regions In SARS-CoV-2-infected cells, exogenous expression A3A but not other APOBEC proteins induced mutations (vRNA). Additionally, mutated C bases were often located at tips bulge or loop vRNA secondary structure. Interestingly, mRNA drastically increased by interferons (IFNs) and tumour necrosis factor-α (TNF-α) epithelial cells derived from respiratory system, site efficient replication. Moreover, rate produced lung treated IFN-ß TNF-α, CRISPR/Cas9-based knockout cells. Collectively, these findings demonstrate is primary host factor drives via editing.

Abstract Emerging in December 2019, coronavirus disease 2019 (COVID-19) eventually became a pandemic and has posed tremendous threat to global public health. However, the origins of SARS-CoV-2, causative agent COVID-19, remain be determined. It reported that certain number early case clusters had contact history with Huanan Seafood Market. Therefore, surveillance SARS-CoV-2 within market is vital importance. Herein, we presented detection results 1380 samples collected from environment animals 2020. By SARS-CoV-2-specific RT-qPCR, 73 environmental tested positive for three live viruses were successfully isolated. The shared nucleotide identity 99.980% 99.993% human isolate HCoV/Wuhan/IVDC-HB-01. In contrast, no virus was detected animal swabs covering 18 species market. SARS-COV-2 nucleic acids showed significant correlation abundance Homo sapiens SARS-CoV-2. summary, this study provided convincing evidence prevalence Market during stage COVID-19 outbreak.

The COVID-19 pandemic has had a significant global impact, with more than 280,000,000 people infected and 5,400,000 deaths. use of personal protective equipment the anti-SARS-CoV-2 vaccination campaigns have reduced infection death rates worldwide. However, recent increase in been observed associated appearance SARS-CoV-2 variants, including recently described lineage B.1.617.2 (Delta variant) B.1.1.529/BA.1 (Omicron variant). These new variants put effectiveness international at risk, outbreaks throughout world. This emergence due to multiple predisposing factors, molecular characteristics virus, geographic environmental conditions, impact social determinants health that favor genetic diversification SARS-CoV-2. We present literature review on most information available analyzed biological, geographical, sociocultural factors development these variants. Finally, we evaluate surveillance strategies for early detection prevent their distribution outside regions.
