Isoquercitrin (quercetin-3-O-β-D-glucopyranoside) has exhibited promising therapeutic potentials as cardioprotective, anti-diabetic, anti-cancer, and anti-viral agents. However, its structural complexity limited natural abundance make both bulk chemical synthesis extraction from medical plants difficult. Microbial biotransformation through heterologous expression of glycosyltransferases offers a safe sustainable route for production. Despite several attempts reported in microbial hosts, the current production levels isoquercitrin still lag behind industrial standards.

Bacillus subtilis is an industrially important microorganism that often used as a microbial cell factory for the production of recombinant proteins due to its food safety, rapid growth, and powerful secretory capacity. However, lack data on functional genes related protein has hindered further development B. factories. Here, strategy combining genome-wide CRISPRi screening targeted CRISPRa activation enhance expression proposed. First, library covering total 4225 coding (99.7%) in genome built corresponding high-throughput methods constructed. Twelve key are identified, including targets without relevant annotations. Meanwhile, transcription by up-regulated. These screened or selected can be easily applied metabolic engineering constructing sgRNA arrays. The relationship between differential pathways engineered strains transcriptome analysis also revealed. High-density fermentation generalisability validation results prove reliability strategy. This method extended other industrial hosts support gene annotation design novel
