Tryptophan is an essential amino acid from dietary proteins. It can be metabolized into different metabolites in both the gut microbiota and tissue cells. such as indole-3-lactate (ILA), indole-3-acrylate (IAC), indole-3-propionate (IPA), indole-3-aldehyde (IAID), indoleacetic (IAA), indole-3-acetaldehyde Kyn produced by intestinal microorganisms through direct Trp transformation also, partly, kynurenine (Kyn) pathway. Abstract Tumor biomarkers, the substances which are produced by tumors or body’s responses to during tumorigenesis and progression, have been demonstrated possess critical encouraging value in screening early diagnosis, prognosis prediction, recurrence detection, therapeutic efficacy monitoring of cancers. Over past decades, continuous progress has made exploring discovering novel, sensitive, specific, accurate tumor significantly promoted personalized medicine improved outcomes cancer patients, especially advances molecular biology technologies developed for detection biomarkers. Herein, we summarize discovery development including history conventional innovative used biomarker classification biomarkers based on tissue origins, application clinical management. In particular, highlight recent advancements biomarker-based anticancer-targeted therapies emerging as breakthroughs promising strategies. We also discuss limitations challenges that need be addressed provide insights perspectives turn into opportunities this field. Collectively, multiple emphasized review may guidance precision medicine, broaden horizons future research directions, expedite patients according their rather than organs origin.

B7-H3 (CD276), a member of the B7 family proteins, is key player in cancer progression. This immune checkpoint molecule selectively expressed both tumor cells and within microenvironment. In addition to its function, has been linked cell proliferation, metastasis, therapeutic resistance. Furthermore, drastic difference protein expression levels between normal tissues suggests that targeting with drugs would lead cancer-specific toxicity, minimizing harm healthy cells. These properties make promising target for therapy.Recently, important advances research drug development have reported, these new findings, including involvement cellular metabolic reprograming, stem enrichment, senescence obesity, expanded our knowledge understanding this molecule, which guiding future strategies B7-H3. review, we briefly discuss biology function development. We emphasize more on latest findings their underlying mechanisms reflect research. addition, improvements B-H3 inhibitors

Tumor microbiota can produce active metabolites that affect cancer and immune cell signaling, metabolism, proliferation. Here, we explore tumor gut microbiome features chemoradiation response in patients with cervical using a combined approach of deep sequencing, targeted bacterial culture, vitro assays. We identify an obligate L-lactate-producing lactic acid bacterium found tumors, Lactobacillus iners, is associated decreased survival patients, induces chemotherapy radiation resistance cells, leads to metabolic rewiring, or alterations multiple pathways, tumors. Genomically similar bacteria commensal other body sites are also significantly colorectal, lung, head neck, skin cancers. Our findings demonstrate the microenvironment alter metabolism lactate signaling causing therapeutic resistance. Lactic could be promising targets across types.

Microbiota-tumor interactions have qualified microbiota as a promising prognostic biomarker in various types of cancers. Although the nasopharynx acts crucial niche upper respiratory tract microbiome, whether intratumoral exists and its clinical significance nasopharyngeal carcinoma (NPC) remain uncertain. To evaluate for individual prognostication patients with NPC.This retrospective cohort study included NPC biopsy samples from 2 hospitals: Sun Yat-sen University Cancer Center (Guangzhou, China) Zhejiang Hospital (Hangzhou, between January 2004 November 2016, follow-up through 2020. A total 802 were according to following criteria: histologically proven NPC, without distant metastasis at initial diagnosis, had not received antitumor treatment before sampling, aged 18 70 years, complete medical records regular follow-up, history cancer, successfully extracted enough DNA experiments. The primary end point was disease-free survival, secondary points metastasis-free survival overall survival. To assess existence load 96 or tumor relapse, 16S rRNA sequencing quantitative polymerase chain reaction used. The associations bacterial outcome evaluated 241 fresh-frozen (training cohort) validated paraffin-embedded internal (n = 233) external 232) validation cohorts. Metagenomic transcriptome analyses performed ascertain origin underlying mechanism bacteria.A (mean [SD] age, 46.2 [10.6] years; 594 [74.1%] male) enrolled. Microbiota presented within tissues, among which Corynebacterium Staphylococcus predominated. Patients high training inferior rates (hazard ratio [HR], 2.90; 95% CI, 1.72-4.90; P < .001), (HR, 3.18; 1.58-6.39; 3.41; 1.90-6.11, .001) than those low load, finding that by Single-nucleotide variant analysis revealed main bacteria. Transcriptome digital pathology demonstrated higher negatively associated T-lymphocyte infiltration.Intratumoral robust tool this study, indicating potential guidance decisions different levels risk malignant progression.
