The SARS-CoV-2 pandemic has led to an urgent need for effective and rapid diagnostic tools. In the present study, we have evaluated predictive potential of nasal microbiota by analyzing microbial community structures at different taxonomic level resolutions—species, genus, family, order, class phylum—using Random Forest modelling. A total 179 swabs from COVID-19-positive (n = 85) COVID-19-negative 94) individuals were sequenced using a full-length 16S rRNA sequencing (Oxford Nanopore) approach. During each iteration model, dataset was randomly split into training set (70%) testing (30%). Model performance improved with finer resolution, achieving highest accuracy Species (AUROC 0.821 ± 0.059) sensitivity 55.6% specificity threshold 90%. progressive decline in AUROC observed broader levels. Here, 1,375 fecal metagenomes from six in-house and published datasets were analyzed, including 128 right-sided CRC (rCRC), 168 left-sided (lCRC), 250 rectal (RC), 829 controls.
