Trigeminal neuralgia is an incurable progressive nervous system disease that can last for several months or years. Stem cells from human exfoliated deciduous teeth (SHED) are a candidate source cell-based therapy. Owing to their neuroprotective and immunomodulatory effects, these neural crest have potential roles in mediating chronic pain. In this study, we established rat model of constriction injury the infraorbital nerve (CCI-ION) evaluate analgesic effect SHED neuropathic The effects local transplantation on inflammatory cell infiltration trigeminal were investigated based hematoxylin eosin staining. levels proinflammatory factors injured transient receptor vanilloid type 1 (TRPV1) expression ganglion quantified. data showed systemic injection attenuated sensitivity rats mechanical stimuli after injury, lasted throughout observation period 8 weeks. PKH26-labeled distributed ipsilateral ganglions 24 72 hours injection. at lesion site led reduced cytokine inhibited CCI-ION-induced upregulation TRPV1 early phase. Therefore, results provide preclinical evidence supports use treatment potentially other pain conditions.

Abstract Peripheral tissue damage and associated inflammation can trigger neuroplastic changes in somatic pain pathways, such as reduced neuronal firing thresholds synaptic potentiation, that ultimately lead to peripheral sensitization chronic pain. Electroacupuncture (EA) relieve inflammatory pain, but the underlying mechanisms are unknown, including contributions of higher centers somatosensory cortex (SSC). We investigated these using optogenetic modulation SSC activity a mouse model. Injection Complete Freund's Adjuvant into hind paw reliably induced accompanied by mechanical thermal (hyperalgesia) within three days (mechanical: 1.54 ± 0.13 g; thermal: 3.94 0.43 s). Application EA produced significant analgesia, responses were reversed activation neurons, suggesting EA-induced analgesia involves central pathways. Western blot immunostaining revealed also attenuated CaMKIIα signaling dorsal root ganglion, spinal cord, SSC, anterior cingulate (ACC). In contrast, ACC. These findings suggest AE suppressing CaMKIIα-dependent plasticity cortical The ACC pathways may be valuable therapeutic targets for treatment.

Chemotherapy-induced peripheral neuropathy is a common factor in limiting therapy which can result cessation or dose reduction. Gabapentin, calcium channel inhibitor, and duloxetine, serotonin noradrenaline reuptake are used to treat variety of pain conditions such as chronic low back pain, postherpetic neuralgia, diabetic neuropathy. It has been reported that administration gabapentin suppressed oxaliplatin- paclitaxel-induced mechanical hyperalgesia rats. Moreover, duloxetine shown suppress oxaliplatin-induced cold allodynia However, the mechanisms by these drugs prevent remain unknown. Behavioral assays were performed using plate von Frey test. The expression levels proteins examined western blot analysis. In this study, we investigated mice. We found prevented development allodynia. addition, our results revealed extracellular signal-regulated protein kinase 1/2 (ERK1/2) phosphorylation spinal cord PD0325901 neuropathic-like behavior inhibiting ERK1/2 activation summary, findings suggest gabapentin, Therefore, could be an effective preventive strategy against
