This study assessed the possible protective role of green synthesized zinc oxide nanoparticles using Moringa olifera leaf extract (MO-ZNPs) in acrylamide (ACR)-induced reproductive dysfunctions male rats. ACR (20 mg/kg b.wt/day) and/or MO-ZNPs (10 were given orally by gastric gavage for 60 days. Then, sperm parameters; testicular enzymes; oxidative stress markers; hormones including testosterone, luteinizing hormone (LH)-estradiol, and follicle-stimulating (FSH) concentration; testis histology; steroidogenesis-related gene expression; apoptotic markers examined. The findings revealed that significantly ameliorated ACR-induced decline gonadosomatic index altered pituitary-gonadal axis, reflected decreased serum testosterone FSH with increased estradiol LH, analysis disruption. Furthermore, a notable restoration tissue content antioxidants (catalase reduced glutathione) but depletion malondialdehyde was evident MO-ZNPs+ACR-treated rats compared to ACR-exposed ones. In addition, oral dosing markedly rescued histopathological changes caspase-3 reactions resulting from exposure. rats, downregulation steroidogenesis genes proliferating cell nuclear antigen (PCNA) immune-expression reversed. Conclusively, protected toxicity suppressing injury apoptosis while boosting sex hormones.

Active vitamin D analog eldecalcitol is clinically applied in treatment of postmenopausal osteoporosis. This study aims to determine the role protection osteocytes from senescence and associated ferroptosis. The MLO-Y4 were exposed D-gal inducing senescence. ovariectomized (OVX) mice treated with using as an aging inducer intraperitoneally injected eldecalcitol. multiplexed confocal imaging, fluorescence situ hybridization transmission electron microscopy assessing osteocytic properties. Immunochemical staining immunoblotting carried out detect abundance expression molecules. ablation receptor led a reduction amounts osteocytes, loss dendrites, increase mRNA SASP factors protein senescent factors, well changes ferroptosis-related genes (PTGS2 & RGS4). Eldecalcitol reversed phenotypes cells shown by improving cell morphology density, decreasing β-gal-positive accumulation, down-regulating (P16, P21 P53). reduced intracellular ROS MDA productions, elevated JC-1 aggregates, up-regulated Nrf2 GPX4. exhibited osteopreserve effects D-gal-induced OVX mice. imaging displayed its improvement on network organization. decreased numbers at tibial diaphysis SADS assay attenuated down-regulated senescence-related restored levels ferroptotic biomarkers osteocytes-enriched bone fraction. It 4-HNE area, stimulated Nrf2-positive staining, promoted nuclear translocation inhibited Nrf2, respectively, present revealed ameliorative ferroptosis contributing preservation against osteoporosis

The evolutionary theory of aging supports a trade-off relationship between reproduction and aging. Aging the male reproductive system primarily affects testes, leading to decrease in levels sexual hormones, alterations sperm quality production, decline fertility that does not necessarily involve complete cessation spermatogenesis. Inflammation, oxidation, apoptosis are events considered as predictors pathogenesis development age-related diseases frequently observed aged testes. Although molecular mechanisms still poorly understood, accumulating evidence points toward pro-inflammatory molecules reactive oxygen species primary contributing factors for testicular However, real impact aging-related on fertility, health, life span is far from being fully revealed. This work discusses current knowledge testis, particularly dysregulated inflammation oxidative damage functioning its different cell populations. More interestingly, this review covers potential benefits anti-aging interventions therapies using either pharmacological compounds (such non-selective non-steroidal anti-inflammatory medication) or more natural alternatives various nutraceuticals even probiotics) exhibit anti-inflammatory, antioxidant, anti-apoptotic properties. Some these currently investigated already clinical use delay prevent

Testicular torsion (TT) is the most common urological emergency in children and young adults that can lead to infertility many cases. The ischemia-reperfusion (IR) injury due TT has been implicated pathogenesis of testicular damage. main pathological mechanisms contralateral after ipsilateral are not fully understood. In presented study, we investigated molecular microscopic basis following detorsion (T/D) explored possible protective role vitamin D3. biochemical analysis indicated IR T/D significantly decreased activity glutathione peroxidase (GPx) enzyme, level serum testosterone, inhibin B, expression miRNA145, while increased myeloperoxidase (MPO) malondialdehyde (MDA), antisperm-antibody (AsAb), ADAM-17. histological semen revealed testis caused damages on different tissues testis. Interestingly, administration D3 prior reversed deterioration effect as by which normal appearance seminiferous tubules with an apparent decrease collagen fiber deposition both testes. Our results treatment could be attributed target miRNA145 ADAM17 protein. To further investigate these findings, performed a detailed modelling study order explore binding affinity toward ability bind active site protein via set hydrophobic hydrophilic interactions high docking score. conclusion, this highlights pharmacological application ameliorate targeting

Aging is a natural and inescapable phenomenon characterized by progressive deterioration of physiological functions, leading to increased vulnerability chronic diseases death. With economic medical development, the elderly population gradually increasing, which poses great burden society, economy field. Thus, healthy aging has now become common aspiration among people over world. Accumulating evidence indicates that substances can mediate deteriorated processes are highly likely have potential prolong lifespan improve aging-associated diseases. Foods from sources full bioactive compounds, such as polysaccharides, polyphenols, carotenoids, sterols, terpenoids vitamins. These compounds their derivatives been shown be able delay and/or diseases, thereby prolonging lifespan, via regulation various processes. Here, we summarize current understanding anti-aging activities vitamins food sources, modes action in delaying improving This will certainly provide reference for further research on effects sources.
