Poria cocos (P. cocos) has been traditionally used as folk medicine and functional food in China for more than 2000 years. The water-soluble polysaccharide is the main component of P. decoction. effects mechanisms from (PCWP) were investigated chronic sleep deprivation (CSD)-induced anxiety rats. CSD induced anxiety, gut dysbiosis, inflammatory responses, reduced neurotransmitter levels, whereas PCWP intervention ameliorated anxiety-like behaviors, increased levels 5-hydroxytryptamine, dopamine, norepinephrine, γ-aminobutyric acid hypothalamus, regulated gastrointestinal peptide factors, inhibited tumor necrosis factor (TNF)-α/nuclear (NF)-κB signaling pathway rats with CSD. changes intestinal flora composition determined using 16S rDNA sequencing, indicated that significantly improved species richness diversity adjusted abundance following dysregulated bacteria closer to normal group: Rikenellaceae_RC9_gut_group, Ruminococcus, Prevotellaceae_UCG-001, Prevotellaceae_NK3B31_group, Fusicatenibacter. Metabolomics was analyze fecal samples identify altered metabolites PCWP-treated groups. Thirty-eight PCWP-related four metabolic pathways such sphingolipid metabolism, taurine hypotaurine vitamin B6 glycerophospholipid metabolism explored. results serum metabolomics showed 26 biomarkers changed after compared model group. regulatory enrichment on sphingolipid, phenylalanine, validation activity enzymes involved above pathways. A strong correlation between potential found. Our findings present new evidence supporting effect preventing progression by inhibiting TNF-α/NF-κB pathway, alleviating disorders, ameliorating microflora imbalance.

Liver disease has become an important risk factor for global health. Resveratrol (Res) is a natural polyphenol which widely found in foods and variety of biological activities. This study investigated the role microbiota-gut-liver axis Res relieving liver fibrosis induced by inorganic mercury exposure. Twenty-eight mice were divided into four groups (n = 7) treated with mercuric chloride and/or 24 weeks, respectively. The results showed that mitigated ileum injury restrained LPS alcohol entering body circulation. Network pharmacological molecular analyses alleviated oxidative stress, metabolism disorders, inflammation, hepatic stellate cell activation liver. In conclusion, alleviates via activating Sirt1/PGC-1α signaling pathway regulating microbial-gut-liver axis, particularly, increasing relative enrichment Bifidobacterium intestinal tract.
