Carbamate is an emerging class of a polymer backbone for constructing sequence-defined, abiotic polymers. It expected that new functional materials can be de novo designed by controlling the primary polycarbamate sequence. While amino acids have been actively studied as building blocks protein folding and peptide self-assembly, carbamates not widely investigated from this perspective. Here, we combined infrared (IR), vibrational circular dichroism (VCD), nuclear magnetic resonance (NMR) spectroscopy with density theory (DFT) calculations to understand conformation carbamate monomer units in nonpolar, aprotic environment (chloroform). Compared acid blocks, are more rigid, presumably due extended delocalization π-electrons on backbones. Cis configurations amide bond energetically stable carbamates, whereas peptides often assume trans at low energies. This study lays essential foundation future developments carbamate-based sequence-defined material design.

Following the successful introduction of two benign solvents for cleaving protected acid peptide fragments from 2-chlorotrityl chloride (2-CTC) resin, there is a need to green cleavage process obtaining amide fragments. In this work, p-xylene and toluene are introduced as greener alternates dichloromethane (DCM) preparing Sieber resin. The N-dealkylation demanding chemical reaction, requiring that protocol be optimised ensure complete After 30 min reaction time, only 66 % final product was retrieved even with conventional solvent.
