Pyrrolizidine is among the saturated N-heterocyclic scaffolds most frequently found in natural products and pharmaceutically relevant substances. Herein, a strategy for synthesis of polysubstituted pyrrolizidine-3-ones by catalytic reductive domino-type recyclization properly functionalized isoxazoline N-oxides was developed. The process diastereoselective, one diastereomer (out four possible ones) predominant many studied cases. Using developed method, modifications potent GSK's PDE4 inhibitor MSD's hNK1 antagonist were prepared.

In the past several decades, C-S bond cleavage for construction C-C C-heteroatom has received tremendous attention in chemistry. Although significant progress been made field transition metal strategies, variety novel transition-metal-free strategies have also developed using halogenated reagents, oxidants, acids, bases. Moreover, photochemical electrochemical methods to achieve organosulfur compounds. To date, however, no comprehensive review reported. Therefore, we herein provide major advances compounds, including thioethers, sulfoxides, sulfones, thioacetals, sulfonium salts, sulfur ylides.

An ammonium ylide-based relay annulation was disclosed, which uses DABCO as the catalyst and oxindole-derived α,β-unsaturated ketimines γ-bromo-crotonates starting materials. This method enables rapid assembly of a series structurally novel spiro-polycyclic oxindoles containing bicyclo[4.1.0]heptane moiety through simultaneous generation three new bonds two rings in one step under mild reaction conditions.

Abstract 2‐Pyrazolines are important five‐membered heterocycles, containing two adjacent nitrogen atoms. They can incorporate various functional groups and possess unique biological properties. Moreover, they have been used as useful building blocks. In recent years, the field of 2‐pyrazoline synthesis has experienced enormous progress. Novel strategies developed, many studies reporting improvements or adaptations existing protocols published, giving access to new functionalizations, unexplored structures, and/or enantioselective synthesis. This review summarizes progress made in synthetic methodologies during period from 2012 2022. The functionalization challenges capabilities, scopes limitations reactions, substitution patterns, mechanisms where appropriate, will be discussed.

We present here an iridium-catalyzed diastereo- and enantioselective [4 + 1] cycloaddition reaction of hydroxyallyl anilines with sulfoxonium ylides under mild conditions, leading to 3-vinyl indolines in moderate good yields excellent enantioselectivities. Control experiments disclosed a plausible mechanism.

A mild and efficient protocol for the perhaloacylation of α-carbonyl sulfoxonium ylides has been developed. The commercially available perfluoro- perchloroacid anhydrides were used as acylating agents in catalyst- additive-free reactions to access α-carbonyl-α'-perhaloacyl high yields. reaction offers a simple method prepare valuable polyfluorinated organosulfur molecules.
