Abstract Toll-like receptor 9 (TLR-9) is a protein that helps our immune system identify specific DNA types. Synthetic oligodeoxynucleotides (ODNs) containing unmethylated cytosine-phosphate-guanine (CpG) motifs (CpG-ODNs) are ligand molecules for Toll-like receptor 9 (TLR9), which is expressed by odontoblasts in vitro and dental pulp cells. This study determined the effects of CpG-ODNs on pulpal immunomodulatory response repair following injury. Briefly, upper right first molars three-week-old mice were extracted, immersed Type A (D35) or B (K3) CpG-ODN solutions (0.1 0.8 mM) 30 min, then replanted. Pulpal healing activity assessed hematoxylin-eosin AZAN staining, as well immunohistochemistry. One week operation, inflammatory reactions occurred all experimental groups; however, re-revascularization newly formed hard tissue deposition observed chamber groups at 2. positive trend expression immune cell markers was toward 0.1 mM. Our data suggest that synthetic low concentrations may evoke a long-lasting macrophage-TLR9-mediated pro-inflammatory, rather than anti-inflammatory, to modulate process formation. Unmethylated cytosine-guanine oligodeoxynucleotides (CpG ODNs) have a storied history as agonists for Toll-like receptor 9 (TLR9). CpG ODNs shown promising antitumor effects in preclinical studies by inducing potent proinflammatory immune responses. However, clinical success has been hindered inconsistent efficacy and immune-related toxicities caused systemic exposure to ODNs. We previously identified that glatiramer acetate (GA), an FDA-approved, lysine-rich polypeptide, could complex class B into cationic nanoparticles which persist at the intratumoral injection site while mitigating induction of cytokines mouse tumor models.
