Aim A comprehensive safety assessment of potential probiotic strains was essential for their application in the food industry. This article systematically evaluated characteristics, whole-genome sequence analysis and Lacticaseibacillus paracasei LC86 casei LC89. Methods Firstly, two lactic acid bacteria selected were identified. Secondly, sequencing performed on LC89, antibiotic resistance, pathogenicity, virulence genes analyzed. We tested various properties strains, included tolerance, cell adhesion, hemolytic activity, catalase gelatin hydrolysis, arginine hydrolysis ability, bile salt capacity, mucin degradation, bioamine, D-/L-lactic production susceptibility, to confirm LC89 both vitro vivo . Additionally, we studied acute toxicity mice through a 14-day oral gavage experiment. Results The identified as genomes devoid virulence, resistance pathogenicity genes. exhibited good tolerance temperature, artificial gastric fluid intestinal fluid; they non-hemolytic, all negative. They capability break down proteins demonstrated sensitivity range antibiotics. LD 50 >2 × 10 CFU/kg. Conclusion experimental results above characteristics indicating candidates probiotics human animal applications.

Background Sleep is essential for physical and mental health. However, stress-related sleep disorders are common in Japan, the gut–brain axis may play a role stress management. This study investigated whether consumption of granola containing multiple prebiotic ingredients could alleviate improve insomnia adults with problems, regardless individual differences gut microbiota. Additionally, we aimed to investigate relationship between changes microbiota observed improvements. Method A single-arm uncontrolled trial was conducted 27 high levels disturbance. The participants consumed 50 g prebiotics-containing daily 8 weeks. Subjective quality assessed using Athens Insomnia Scale, Epworth Oguri-Shirakawa-Azumi Inventory-Middle-aged Aged version (OSA-MA). Results After weeks, subjective scores onset maintenance improved significantly, whereas mood disturbance decreased significantly. analysis showed that relative abundance Bifidobacterium increased, Bacteroides decreased. Correlation suggested significant association increased level reduced ( r = −0.39, p 0.0035) −0.3, 0.026). Conclusion Prebiotics-containing disturbances, which be attributed alterations microbiota, particularly increase abundance.

Background: Several randomized controlled trials (RCTs) have shown conflicting results on cortisol levels following probiotic administration in healthy and diseased populations. Previous analyses were inconclusive due to limited studies, evidence is lacking how these effects vary by health status; region; therapy duration; medications, use of single or multiple strains. Methods: In this systematic review meta-analysis (PROSPERO [CRD42024538539]), we searched PubMed, Cochrane Library, Embase, Scopus, Web Science, CINAHL, ProQuest, Science Preprints until 13 August 2024, for RCTs administration, either alone combined, across all age groups without specific medical condition requirements. We applied random-effects meta-analysis, assessed bias using the RoB 2 tool, evaluated certainty with GRADE. Findings: screened 1739 records retrieved 46 (3516 participants). Probiotics supplementation decreased compared control arm [46 RCTs; SMD: −0.45; 95% CI: −0.83; −0.07; I2: 92.5%, low certainty]. Among various subgroups; probiotics subgroups concomitant medications [37 −0.30; CI [−0.58; −0.03], 88.7%] a strain [30 −0.33; −0.63; −0.028; 88.8%], population [35 SMD:−0.3; −0.58; −0.03; 88.7] Asia region [21 −1.58; 95%]. Interpretation: A level suggests might reduce levels, but more targeted studies are needed identify variables affecting response subgroups.
