Photodynamic therapy with reactive oxygen species production is a prospective treatment to combat cancer stem cells (CSCs). However, the innate drawbacks, including short lifetime and diffusion distance of hypoxia within solid tumors, have become bottlenecks for clinical applications photodynamic therapy. Here, we develop mitochondria-targeting hemicyanine–oleic acid conjugate (CyOA), which can self-assemble into supramolecular nanoparticles (NPs) without any exogenous excipients. CyOA also shown targeting mitochondrial complex II protein succinate dehydrogenase inhibit oxidative phosphorylation reverse tumor hypoxia, resulting in 50.4-fold higher phototoxicity against breast (BCSCs) compared SO 3 -CyOA NPs that cannot target mitochondria. In 4T1 BCSC models, achieve inhibition less lung metastasis nodules clinically used photosensitizer Hiporfin. This study develops self-assembled small molecule serve as both inhibitor eradication CSCs tumors.

The multi-faceted nature of functionalized magnetic nanoparticles (fMNPs) is well-suited for cancer therapy. These nanocomposites can also provide a multimodal platform targeted therapy due to their unique guidance characteristics. When induced by an alternating field (AMF), fMNPs convert the magnetostatic energy heat hyperthermia (MHT), as well controlled drug release. Furthermore, with ability near-infrared (NIR) light energy, have attracted interest photothermal (PTT). Other than MHT and PTT, place in combination therapies, such chemo-MHT, chemo-PTT, chemo-PTT–photodynamic therapy, among others, versatile properties. Thus, this review presents multifunctional based on AMF or NIR light. We will first discuss different chemo-MHT. Secondly, we irradiated lasers PTT chemo-PTT. Finally, used dual-mode + NIR-laser-induced magneto-photo-hyperthermia (MPHT) be discussed.

Abstract This review follows the research, development and clinical applications of photosensitizer 5,10,15,20‐tetra( m ‐hydroxyphenyl)chlorin ( THPC, temoporfin) in photodynamic (cancer) therapy (PDT) other medical applications. Temoporfin is active substance medicinal product Foscan® authorized EU for palliative treatment head neck cancer. Chemistry, biochemistry pharmacology, as well temoporfin are addressed, including extensive work that has been done on formulation liposomal formulations. The literature covered from 2009 to early 2022, thereby connecting it previous this published journal [Senge, M. O. J. C. Brandt (2011) Photochem.
