Deep brain stimulation (DBS) of the nucleus accumbens (NAc) (NAc-DBS) is an effective solution to refractory obsessive-compulsive disorder (OCD). However, evidence for neurobiological mechanisms OCD and effect NAc-DBS still lacking. One hypothesis that electrophysiological activities in NAc are modulated by DBS, another neurotransmitters influenced DBS. To investigate these potential alterations, rats with quinpirole (QNP)- induced were treated DBS core part NAc. Then, extracellular spikes (SPK) local field potentials (LFP) recorded, levels relevant related proteins measured. Analysis SPK revealed firing rate was decreased pattern changed after NAc-DBS, analysis LFP showed overall power spectral density (PSD) reduced NAc-DBS. Additionally, we found relative powers theta band, alpha band beta increased status, while delta gamma decreased. This pathological distribution reformed Furthermore, monoamines [dopamine (DA) serotonin (5-HT)] amino acids [glutamate (Glu) gamma-aminobutyric acid (GABA)] expression two types DA receptors exhibited opposite change. These abnormalities could be reversed findings provide a more comprehensive understanding about function pathophysiology detailed

Itch is an uncomfortable and complex sensation that elicits the desire to scratch. The nucleus accumbens (NAc) activity important in driving sensation, motivation, emotion. Excitatory afferents from medial prefrontal cortex (mPFC), amygdala, hippocampus are crucial tuning of dopamine receptor D1-expressing D2-expressing medium spiny neurons (Drd1-MSN Drd2-MSN) NAc. However, a cell-type neural circuity-based mechanism NAc underlying acute itch remains unclear. We found induced by compound 48/80 (C48/80) decreased intrinsic membrane excitability Drd1-MSNs, but not Drd2-MSNs, core male mice. Chemogenetic activation Drd1-MSNs alleviated C48/80-induced scratching behaviors itch-related anxiety-like behaviors. In addition, C48/80 enhanced frequency spontaneous EPSCs (sEPSCs) reduced paired-pulse ratio (PPR) electrical stimulation-evoked Drd1-MSNs. Furthermore, increased excitatory synaptic mPFC, basolateral amygdala (BLA) or ventral (vHipp).
