Alzheimer’s disease, characterized by a decline in cognitive functions, is frequently associated with decreased levels of acetylcholine due to the overactivity acetylcholinesterase (AChE). Inhibiting AChE has been key therapeutic strategy treating yet search for effective inhibitors, particularly from natural sources, continues their potential fewer side effects. In this context, three new alkaloids—oleracone L, portulacatone B, and portulacatal—extracted Portulaca oleracea L., have recently shown promising anticholinesterase activity vitro. However, no experimental or computational studies explored binding potential. This study represents first comprehensive silico analysis these compounds, employing ADME prediction, molecular docking, dynamics simulations, MM-PBSA calculations assess The drug-likeness was evaluated based on Lipinski, Pfizer, Golden Triangle, GSK rules, all alkaloids meeting criteria. profiles suggested that can effectively cross blood–brain barrier, critical requirement treatment. Molecular docking revealed oleracone L had highest affinity (−10.75 kcal/mol) towards AChE, followed portulacatal demonstrating significant interactions crucial enzyme residues. simulations over 200 ns confirmed stability interactions, RMSD values below 2 Å complexes, indicating stable throughout simulation period. RMSF radius gyration analyses further corroborated minimal impact enzyme’s overall flexibility compactness. Moreover, provided additional support efficacy, showing most favorable energy, could be superior inhibitor, potentially its stronger more consistent hydrogen bonding electrostatic compared other studied alkaloids. These findings highlight efficiency viability as suggesting they candidates disease underscores importance validation through vitro vivo experiments confirm predictions.

Background: In recent decades, the incidence of depression has gradually increased in general population globally. Depression is also common during gestation and could result detrimental gestational complications for both mother fetus. The survey presented aimed to evaluate whether pregnant women’s perinatal be associated with socio-demographic, anthropometry lifestyle factors, postnatal outcomes. Methods: This a cross-sectional conducted on 5314 women. Socio-demographic factors were recorded by relevant questionnaires via face-to-face interviews. Anthropometric parameters measured qualified personnel. Perinatal depressive symptomatology status was evaluated Beck’s Inventory (BDI-II) questionnaire. Results: Depressive symptoms throughout found 35.1% enrolled significantly lower educational economic level, pre-pregnancy regular smoking reduced levels Mediterranean diet adherence levels, higher prevalence diabetes preterm birth, as well delivering caesarean section abnormal childbirth weight. maternal postpartum exclusive breastfeeding practices, childhood asthma. Conclusions: Pregnant appears various anthropometry, characteristics frequency several adverse pregnancy complications. present findings emphasize importance mental health, highlighting need develop apply public strategies policies psychological counseling support future mothers minimize probable risk that may trigger depression. Novel well-organized, follow-up surveys enhanced validity are highly recommended establish more definitive conclusions.

Abstract This research focuses on developing environmentally friendly and economically viable Co/Ni-doped hematite nanoparticles (HNPs) through both chemical green synthesis methods evaluated their potential for biomedical environmental applications. The employs polyvinylpyrrolidone (PVP), while the approach utilizes Azadirachta indica ( A. ) leaf extract as a stabilizing agent. HNPs are crystalline size ranging from 14 to 21 nm, morphology analysis revealed that NPs exhibited quasi-spherical, with an average particle 15.98 25.91 dopants confirmed contain by XPS spectra. VSM study explains magnetic parameters, coactivity, residual magnetism, magnetization. plants quinones, saponins, glycosides, alkaloids, flavonoids. Characterization of reveals optimized enhanced photocatalytic activity. These exhibit remarkable 93%–95% degradation UV-reactive dyes (methyl orange methylene blue) within 90 min, attributed structural surface modifications improve light absorption enhance charge separation. concludes green-synthesized outperform chemically synthesized counterparts superior photocatalysts. Additionally, antioxidant evaluations using 2,2-diphenyl-1-picrylhydrazyl (DPPH) nitric oxide (NO) assays suggest significant capabilities. A high scavenging activity percentage, 83% 88%, was observed, which increased higher concentrations making these suitable applications require system. In this study, IC 50 values against DPPH/NO assay were calculated be 18.33 μg ml −1 16.09 , respectively. highlights multifunctional properties HNPs, addressing demand tailored inorganic minimal ecological impact.

Epilepsy affects more than 70 million individuals of all ages worldwide and remains one the most severe chronic noncommunicable neurological diseases globally. Several neurotransmitters, membrane protein channels, receptors, enzymes, and, recently noted, various pathways, such as inflammatory mTORC complexes, play significant roles in initiation propagation seizures. Over past two decades, developments have been made diagnosis treatment epilepsy. Various pharmacological drugs with diverse mechanisms action other options developed to control seizures treat These include surgical treatment, nanomedicine, gene therapy, natural products, nervous stimulation, a ketogenic diet, gut microbiota, etc., which are developmental stages. Despite plethora options, one-third affected resistant current medications, while majority approved side effects, changes can occur, pharmacoresistance, effects on cognition, long-term problems, drug interactions, risks poor adherence, specific for certain psychological complications. Therefore, development new that no or minimal adverse is needed combat this deadly disease. In Review, we comprehensively summarize explain stages epilepsy well their status clinical trials advancements.
