Metabolic dysfunction-associated steatotic liver disease (MASLD) comprises a spectrum of diseases that span simple steatosis, metabolic steatohepatitis (MASH) and fibrosis may progress to cirrhosis cancer. Abstract Background Dolichandrone spathacea (D. spathacea) is a common traditional Chinese medicine extensively used to treat chronic bronchitis (CB). However, phytochemicals and potential mechanisms of this species against CB have not been fully illuminated. Therefore, study aimed isolate elucidate the D. spathacea, clarify its molecular key therapeutic targets in treating through network pharmacology validate these findings using docking, experimental approaches. Results Three compounds, beta-sitosterol, 6-O-trans-p-coumaroyl ajugol, 6-O-[(E)-4-methoxycinnamoyl] catalpol were isolated from EtOAc fraction, with beta-sitosterol being reported for first time species. After combining species identified those literature references, 59 compounds obtained, 30 bioactive screened. Among these, luteolin was predicted interact highest number CB-related proteins. Using GeneCards DrugBank databases, 66 intersecting target genes identified between spathacea CB. The protein-protein interaction analysis core targets, including TNF, AKT1, SRC, EGFR, IL2, MMP-9, HSP90AA1, PTGS2. KEGG enrichment analyses suggested that exerts effects on by modulating various biological processes pathways. Notably, top three – PTGS2, MMP-9 enriched TNF IL-17 signaling pathways. computational docking could bind all spathacea. validation revealed ethanol extract inhibited nitric oxide production induced LPS, an IC50 value 25.34 μg/mL.
