Abstract Circadian disruptions may result in sleep problems, oxidative stress and an altered inflammatory response. These symptoms contribute to PD pathogenesis, despite a lack of direct experimental evidence supporting this relationship. Clock genes are essential drive maintain circadian rhythm. To elucidate the possible role PD, we investigated 132 tag variants eight clock genes. We genotyped these tags within 1,394 Chinese cases 1,342 controls using Illumina GoldenGate chips. discovered that SNPs ARNTL (rs900147, P = 3.33 × 10 −5 , OR 0.80) PER1 (rs2253820, 5.30 −6 1.31) significantly associated with risk. Moreover, positive association rs900147 variant was more robust tremor dominant (TD) ( 3.44 −4 ) than postural instability gait difficulty (PIGD) 6.06 −2 ). The rs2253820 PIGD 5.42 TD 4.2 Haplotype analysis also showed were PD. Imputation identified some which affect gene expression through altering transcription factor binding site. In summary, our findings suggest genetic polymorphisms genes, as well disruptions, pathogenesis.

Over the last decade, a dual character of cell response to oxidative stress, eustress versus distress, has become increasingly recognized. A growing body evidence indicates that under physiological conditions, low concentrations reactive oxygen and nitrogen species (RONS) maintained by activity endogenous antioxidant system (AOS) allow reversible oxidative/nitrosative modifications key redox-sensitive residues in regulatory proteins. Epidemiological studies have provided evidence that traffic noise exposure is linked to cardiovascular diseases such as arterial hypertension, myocardial infarction, and stroke. Noise a nonspecific stressor activates the autonomous nervous system endocrine signaling. According reaction model introduced by Babisch colleagues, chronic low levels of can cause so-called nonauditory effects, disturbances activity, sleep, communication, which trigger number emotional responses, including annoyance subsequent stress. Chronic stress in turn associated with risk factors, comprising increased blood pressure dyslipidemia, viscosity glucose, activation clotting animal models humans. Exposure to traffic noise is associated with stress and sleep disturbances. The World Health Organization (WHO) recently concluded that road increases the risk for ischemic heart disease potentially other cardiometabolic diseases, including stroke, obesity, diabetes. WHO report focused on whole-day exposure, but new epidemiological translational field studies indicate nighttime noise, in particular,is an important factor cardiovascular (CVD) through increased levels of hormones vascular oxidative stress, leading endothelial dysfunction subsequent development various CVDs. Novel experimental found be stress–induced brain damage, mediated by activation NADPH oxidase, uncoupling neuronal nitric oxide synthase, vascular/brain infiltration inflammatory cells. Noise-induced pathophysiology was more pronounced response as compared daytime noise. This review focuses consequences

Cardiovascular disease, which can lead to angina and shortness of breath, remains one the most serious threats human health. Owing its imperceptible symptoms, it is difficult determine pathogenesis treatment methods for cardiovascular disease. Nuclear factor erythropoietin-2-related 2/heme oxygenase 1 (Nrf2/HO-1) a protein found in all cells body. It activated, transferred nucleus, bound DNA by antioxidant response elements (AREs). As regulator system, upregulates expression HO-1 reduce oxidative stress. Nrf2/HO-1 also has ability modulate calcium levels prevent ferroptosis, pyroptosis, autophagy, programmed cell necrosis, alkaliptosis, clockophagy. In view importance regulation homeostasis, this review summarizes current research on relationship between disease Nrf2/HO-1. Normal diseases, such as viral myocarditis myocardial ischemia-reperfusion injury, have been treated with Rheumatic heart cardiac tumors, arteriosclerosis, arrhythmia, hypertensive infarction during experiments. Research demonstrated clinical application pediatric disease; further trials will help elucidate potential signaling axis.
