Per- and polyfluoroalkyl substances (PFAS) are diverse synthetic chemicals manufactured over seven decades. It is an aliphatic molecule with a basic hydrophobic structure of carbon fluorine linked to hydrophilic end group. Due their physicochemical properties associated the unique structure, PFAS has been used in wide variety applications including aqueous film-forming foams (AFFF), paper, carpets, non-stick cookware, etc. as they make products resistant water, heat, stains. These molecules have drawn great attention recently for properties, high stability low degradability, so-called “Forever Chemicals”. strongest carbon-fluorine bond which makes them persistent environment. Hence it contaminates natural resources endangers public health. Polyfluoroalkyl chemicals (PFCs) present potential health risks due to their persistence and bioaccumulation. However, there is currently insufficient evidence regarding impact on cardiovascular disease (CVD). Consequently, it imperative investigate the correlation between PFCs CVD. The data was collected from National Health Nutrition Examination Survey in 2005-2012. Logistic regression models were employed assess association single PFC Generalized additive model (GAM) used for evaluating nonlinear relationships. Subgroup analyses conducted explore interaction effects. Bayesian kernel machine (BKMR) weighted quantile sum (WQS) evaluate joint effect of exposures In logistic regression, PFDE, MPAH, PFUA positively associated with GAM, a significant relationship MPAH analysis revealed gender race effects correlated CVD males but show no difference females. Perfluorooctanesulfonate (PFOS), an emerging contaminant with widespread concern, has been associated the pathogenesis of atherosclerosis (AS). As a substitute for PFOS, sodium p-perfluorous nonenoxybenzenesulfonate (OBS) is extensively utilized in various applications and detected human blood. However, its potential health risk AS remain unclear. In this study, we investigated comparative impacts PFOS OBS on endothelial dysfunction atherogenesis. vivo Apolipoprotein E knockout (ApoE-/-) mice were exposed to 0.4 or 4 mg/L PFOS/OBS 12 weeks. We found that dyslipidemia developed more rapidly OBS-exposed than PFOS-exposed mice. exhibited higher enrichment capacity both blood aortic tissues OBS. Remarkably, induced pronounced inflammatory response caused significant disruption barrier aorta ApoE-/- compared PFOS. vitro experiments showed OBS, at same exposure concentrations durations as (0.1-20 μmol/L, 48 h), effectively inhibited cell viability umbilical vein cells (HUVECs), levels lactate dehydrogenase (LDH) release, enhanced adhesion between HUVECs monocytes. Both activate NF-κB signaling pathway upregulate expression factors. Notably, use but not was shown disrupt junctions increase permeability by activating MAPK/ERK pathway. Background Studies have shown that ESCRT genes affect cell aging, immune environment, and tumors. BRCA was used to explore its effects on tumor prognosis therapy. Methods The data sets of Cancer Genome Atlas (TCGA), Genome-Tissue Expression Plan (GTEX), Human Protein Mapping (HPA), Gene Omnibus (GEO), Clinical Proteomic Tumor Analysis Consortium (CPTAC), R software package, bioinformatics methods were mine the potential carcinogenic ESCRT, including expression level, prognostic value, value in various types tissues, function family further verified breast cancer.
