Abstract Osteoporosis (OP) can result in slower bone regeneration than the normal condition due to abnormal oxidative stress and high levels of reactive oxygen species (ROS), a detrimental for formation, making OP‐related healing significant clinical challenge. As osteogenic differentiation ability marrow mesenchymal stem cells (BMSCs) is closely related regeneration; currently, this study assessed effects Picein on BMSCs vitro osteoporotic defect vivo. Cell viability was determined by CCK‐8 assay. The production malonaldehyde, superoxide dismutase activities, glutathione evaluated using commercially available kits, flow cytometry analysis adopted detect macrophage polarization. Osteogenic capacity alkaline phosphatase (ALP) activity, ALP staining, Alizarin red S staining. expression osteogenic‐related proteins (OPN, Runx‐2, OCN) genes (ALP, BMP‐4, COL‐1, Osterix) were Western blotting real‐time PCR (RT‐PCR). In addition, proliferation, migration ability, angiogenic human umbilical vein endothelial (HUVECs) EdU scratch test, transwell assay, tube formation respectively. Angiogenic‐related (VEGF, vWF, CD31) also RT‐PCR. Results showed that alleviated erastin‐induced stress, enhanced BMSCs, angiogenesis HUVECs, protects against ferroptosis through Nrf2/HO‐1/GPX4 axis. Moreover, regulate immune microenvironment promoting polarization M2 macrophages vitro. reduce inflammation promotes OP rat models Altogether, these results suggested promote alleviate via pathway, offering as novel antioxidant agent treating defect.

The treatment of osteoporotic bone defect remains a big clinical challenge because osteoporosis (OP) is associated with oxidative stress and high levels reactive oxygen species (ROS), condition detrimental for formation. Anti-oxidative nanomaterials such as selenium nanoparticles (SeNPs) have positive effect on osteogenesis owing to their pleiotropic pharmacological activity which can exert anti-oxidative functions prevent loss facilitate regeneration in OP. In the current study strategy one-pot method by introducing Poly (lactic acid-carbonate) (PDT) β-Tricalcium Phosphate (β-TCP) SeNPs, developed prepare an injectable, anti-collapse, shape-adaptive adhesive graft substitute material (PDT-TCP-SE). PDT-TCP-SE exhibits sufficient adhesion biological microenvironments osteoinductive activity, angiogenic anti-inflammatory well vitro vivo. Moreover, protect BMSCs from erastin-induced ferroptosis through Sirt1/Nrf2/GPX4 antioxidant pathway, which, together, demonstrated emerging biomaterial potential application future defects. Injectable, adhesive, plastic bioactive was successfully synthesized. Incorporation SeNPs PDT into β-TCP regenerated new in-situ moderating defects area. reduced ROS microenvironment. could also moderate inhibit via pathway vitro. alleviate inflammatory environment promote This has advantages simple synthesis, biocompatibility, regulation level, value tissue engineering.

Introduction According to reports, obesity has a significant impact on bone health. And the weight-adjusted-waist index (WWI), superior BMI and waist circumference (WC), is new indicator arising in recent years. This research investigated relationship between WWI total mineral density (BMD) for adults aged 20 59. Methods Using data from 2011–2018 NHANES, we looked into independent link BMD as well its nonlinearity using weighted multiple linear regression smooth curve fitting. Two-stage models were employed calculate threshold effects. There additional subgroup analyses testing interactions. Results Multiple studies of 10,372 individuals showed inverse 59 [β = -0.04, 95% CI: (-0.04, -0.03), P<0.0001]. smoking, race, chronic kidney disease (CKD) had no effect this negative connection (P interaction >0.05). In addition, found nonlinear diabetic CKD patients, which saturation point was 11.38 cm/√kg patient group 10.29 group. Conclusion Our analysis demonstrated association 20-59.

Abstract Purpose Previous studies have suggested that obesity defined by body mass index(BMI) is a protective factor for bone mineral density(BMD), but overlooked the potential influence of different types obesity. This study aims to evaluate correlation between abdominal index A Body Shape Index(ABSI) and adolescent density, analyze relationship metabolism. Methods total 1557 participants were included in NHANES from 2007 2018. Calculate ABSI using specific formula takes into account waist circumference BMI. weighted multiple linear regression model used BMD. Forest plots are correlations subgroups, cubic splines limited nonlinear saturation effects Results After adjusting confounding factors, there was significant ( P < 0.01) femoral BMD, both as continuous variable an ordered categorical variable. The restrictive spline curve indicates effect Conclusion Research has shown negative BMD at four detection sites femur, this may vary slightly due age, race, family income, sites. research results indicate compared overall weight, fat distribution content be more closely related

Sepsis is a life-threatening organ dysfunction caused by dysregulated host response to infection. The inflammatory cytokine storm causes systemic damage, especially acute lung injury in sepsis. In this study, we found that the expression of S-phase kinase-associated protein 2 (Skp2) was significantly decreased sepsis-induced (ALI). activated MEK/ERK pathway and inhibited Skp2 pulmonary epithelium, resulting reduction K48 ubiquitination solute carrier family 3 member (SLC3A2), thereby impairing its membrane localization cystine/glutamate exchange function. Consequently, intracellular redox reactions induced ferroptosis epithelial cells, leading injury. Finally, demonstrated intravenous administration mRNA-encapsulating lipid nanoparticles (LNPs) epithelium alleviated septic mice. Taken together, these data provide an innovative understanding underlying mechanisms ALI promising therapeutic strategy for
