Abstract
Hepatocellular
carcinoma
(HCC)
is
a
malignant
tumor
of
the
digestive
system
and
one
leading
causes
cancer
mortality
worldwide.
So
far,
various
genetic
mutations
have
been
observed
in
hepatocellular
related
with
tumorigenesis
due
to
genomic
instability.
However,
clinical
characteristic
therapeutic
value
HCC
were
extent
unknown.
In
present
study,
by
analyzing
gene
mutation
landscape
tissues
from
TCGA
database,
we
demonstrated
that
TTN
(titin)
was
high-frequency
HCC,
relevant
poor
prognosis
HCC.
had
no
relationship
immune
signature
but
linked
change
drug
sensitivity
GSK1904529A,
nilotinib,
5-Fluorouracil
(5-FU)
sapitinib.
Furthermore,
through
performing
TIDE
algorithm,
CCK-8
assays,
colony
formation
cell
sphere-forming
experiment,
vivo
transplantation
experiment
biological
experiments
investigated

between
sensitivity.
We
found
led
increased
stability
protein,
which
depletion
results
decrease
intracellular
ferrous
ion
content,
inhibiting
ferroptosis
pathway,
significantly
reduced
5-FU
vitro
vivo.
These
findings
cause
reduce
5-FU,
reducing
treatment
efficacy.
conclusion,
our
studies
provided
initial
evidences
can
sever
as
biomarker
for
predicting
prognosis,
signature,
molecular
targets
therapy
