Abstract Background Parkinson’s disease (PD) is frequently accompanied by mood and chronic fatigue syndrome (CFS) symptoms. It unknown whether immune activation insulin resistance (IR) or brain injuries impacts the severity of affective CFS symptoms due to PD. Aims To examine immune, IR, and/or injury biomarkers determine Methods Using a case (70 PD patients) control (60 healthy controls) study design, we assessed symptoms, measured peripheral immune-inflammatory response system (IRS) using interleukin-6 (IL-6), IL-10, zinc, calcium levels, Homeostasis Model Assessment 2 (HOMA2IR) index, serum markers including S100 calcium-binding protein B (S100B), neuron-specific enolase (NSE), phosphorylated tau217 (pTau217), glial fibrillary acidic (GFAP). Results patients showed increased scores, IRS activation, HOMA2IR, NSE, GFAP, pTau217, S100B levels as compared controls. A large part (52.5%) variance in mood+CFS score was explained regression on S100B, HOMA2IR interleukin-10 (IL-10) (all positively) (inversely). Abstract Background Autoimmune responses contribute to the pathophysiology of Long COVID, affective symptoms and myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS). Objectives To examine whether its accompanying CFS are associated with immunoglobulin (Ig)A/IgM/IgG directed at neuronal proteins including myelin basic protein (MBP), oligodendrocyte glycoprotein (MOG), synapsin, α+β-tubulin, neurofilament (NFP), cerebellar protein-2 (CP2), blood-brain-barrier-brain-damage (BBD) claudin-5 S100B. Methods IgA / IgM/IgG above proteins, human herpes virus-6 (HHV-6) Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) were measured in 90 COVID patients healthy controls, while C-reactive (CRP), advanced oxidation products (AOPP) association ratings additionally assessed a subgroup thereof. Results is significant increases IgG tubulin (IgG-tubulin), MBP, MOG synapsin; IgM-MBP, MOG, CP2, synapsin BBD; IgA-CP2 synapsin. IgM-SARS-CoV-2 IgM-HHV-6 antibody titers significantly correlated IgA/IgG/IgM-tubulin -CP2, IgG/IgM-BBD, IgM-MOG, IgA/IgM-NFP, IgG/IgM-synapsin. Binary logistic regression analysis shows that IgM-MBP IgG-MBP best predictors COVID. Multiple IgG-MOG, CRP AOPP explain together 41.7% variance severity CFS. Neural network IgM-synapsin, IgA-MBP, IgA-synapsin, IgA-CP2, most important due predictive accuracy r=0.801. Conclusion Brain-targeted autoimmunity contributes pathogenesis physio-affective phenome.

Four decades of research in the field Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS) have yielded lessons that may be instructive for those devising criteria to better comprehend Post-Acute Sequelae SARS CoV-2 Infection (PASC) and Long COVID. For instance, substantial effort has been devoted defining classification systems, operationalizing methods, developing instruments with adequate reliability validity ME/CFS field. The current article provides guidelines a case definition COVID discusses significance psychometric issues criterion variance, including how specify symptoms, develop thresholds, subtypes, exclusionary conditions. could enhance our knowledge pathophysiology, early diagnosis, prognosis, identification effective treatments.

Abstract Background Multiple studies have shown that Long COVID (LC) disease is associated with heightened immune activation, as evidenced by elevated levels of inflammatory mediators. However, there no comprehensive meta-analysis focusing on activation the response system (IRS) and compensatory immunoregulatory (CIRS) along other phenotypes in LC patients. Objectives This designed to explore IRS CIRS profiles patients, individual cytokines, chemokines, growth factors, C-reactive protein (CRP) immune-associated neurotoxicity. Methods To gather relevant for our research, we conducted a thorough search using databases such PubMed, Google Scholar, SciFinder, covering all available literature up December 20th, 2023. Results The current encompassed 82 examined multiple profiles, protein, 58 cytokines/chemokines/growth factors 3836 patients versus 4537 normal controls (NC). showed significant increases IRS/CIRS ratio (standardized mean difference (SMD:0.156, confidence interval (CI): 0.051;0.261), (SMD: 0.345, CI: 0.222;0.468), M1 macrophage 0.421, 0.290;0.551), T helper (Th)1 0.353, 0.189;0.517), Th17 0.492, 0.332;0.651) neurotoxicity 0.327 0.205;0.448).
