Serum biomarkers are promising minimally invasive outcome measures in clinical studies Duchenne muscular dystrophy (DMD). However, strongly associated with progression and predicting performance decline lacking. In this study we aimed to identify serum able predict milestones DMD. Towards aim present a retrospective multi-center cohort including samples data collected research participants DMD as part of natural history at the University Florida (UF) real-world observations Leiden Medical Center (LUMC) between 2009-2022. Abstract Musculoskeletal injuries are a substantial source of global disability through weakness and loss function, which can be attributable, in part, to deficits skeletal muscle quality. Poor quality, resulting from fibrotic pathology or fatty infiltration, strongly predicts lower rates patient recovery following injury higher re‐injury. The cellular sources fibrosis infiltration within mesenchymal fibro‐adipogenic progenitors (FAPs), central effectors support homeostasis, regeneration growth. However, acute chronic musculoskeletal injury, FAPs promote fibro/fatty that is likely limit repair. Given their indispensable role muscle, have emerged as compelling target tissue injury. This review provides insight into the aetiology FAP activity various injuries, addition signalling components effect differentiation. Contrasting with therapeutic potential, disease‐ injury‐specific activation further cements crucial improve function enhance outcomes. image
