Atopic dermatitis (AD) is the most common chronic inflammatory skin disease and presents a major public health problem worldwide. It characterized by recurrent and/or course of lesions with intense pruritus. Its pathophysiologic features include barrier dysfunction, aberrant immune cell infiltration, alterations in microbiome that are associated genetic environmental factors. There complex crosstalk between these components, which primarily mediated cytokines. Epidermal dysfunction hallmark AD caused disruption proteins lipids responsible for establishing barrier. To better define role cytokines stratum corneum lipid abnormalities related to AD, we conducted systematic review biomedical literature PubMed from its inception 5 September 2023. Consistent dominant TH2 skewness seen type 2 were featured prominently as possessing central epidermal skin. The TH1 TH17 also identified affect lipids. Considering broad cytokine dysregulation observed pathophysiology, understanding each will help developing therapeutics best achieve homeostasis patients.

Abstract Background Atopic dermatitis (AD) patients display an altered skin microbiome which may not only be indicator but also a driver of inflammation. We aimed to investigate associations among AD patients' microbiome, clinical data, and response systemic therapy in the TREATgermany registry. Methods Skin swabs 157 were profiled with 16S rRNA gene amplicon sequencing before after 3 months treatment dupilumab or cyclosporine. For comparison, 16s data from 258 population‐based healthy controls used. Disease severity was assessed using established instruments such as Eczema Area Severity Index (EASI). Results confirmed previously shown correlation Staphylococcus aureus abundance bacterial alpha diversity measured by EASI. Therapy Dupilumab shifted community toward pattern seen controls. The relative Staphylococci particular S. significantly decreased on both lesional non‐lesional skin, whereas hominis increased. These changes largely independent degree improvement observed for Conclusions Systemic cyclosporine tends restore indicating potential effects IL‐4RA blockade microbiome.

It is now longer than half a century, humans, animals, and nature of the world are under influence exposure to many newly introduced noxious substances. These exposures nowadays pushing borders be considered as causative or exacerbating factors for chronic disorders including allergic, autoimmune/inflammatory, metabolic diseases. The epithelial linings serve outermost body’s primary physical, chemical, immunological barriers against external stimuli. “epithelial barrier theory” hypothesizes that these diseases aggravated by an ongoing periepithelial inflammation triggered wide range barrier–damaging insults lead “epithelitis” release alarmins. Abstract: Atopic dermatitis (AD) is the most common inflammatory skin disease in children. Children with severe AD have a multidimensional burden characterized by lesions, itching, frequent infections, sleep deprivation, and high rate of comorbidities. These impact mental health overall quality life not only children but also their parents caregivers. There are few effective available treatment options for young that suitable long-term use. Due to adverse effects, practice guidelines consider systemic agents inappropriate this age group, although they still used off-label extreme cases. The biologic dupilumab has recently been approved aged 6– 11 years (EU) moderate-to-severe (USA) AD, offering hope population patients unmet clinical need. purpose review describe needs prior approval summarize existing data supporting dupilumab's safety efficacy these Keywords: atopic dermatitis, children, dupilumab, pediatric,

Seborrheic dermatitis (SD) is a chronic inflammatory skin disease characterized by erythematous papulosquamous lesions in sebum rich areas such as the face and scalp. Its pathogenesis appears multifactorial with disbalanced immune system, Malassezia driven microbial involvement barrier perturbations. Microbial has been well described SD, but remains to be properly elucidated. To determine whether impairment critical factor of inflammation SD alongside dysbiosis, cross-sectional study was performed 37 patients mild-to-moderate facial SD.
