Background Adjuvant pembrolizumab significantly improved recurrence-free survival (RFS) and distant metastasis-free (DMFS) versus placebo in the phase 3 KEYNOTE-716 study of resected stage IIB or IIC melanoma. At prespecified third interim analysis (data cut-off, January 4, 2022), HR for RFS overall population was 0.64 (95% CI, 0.50 to 0.84) DMFS 0.47 0.88). We present a post hoc efficacy by subtypes defined histopathologic characteristics. Methods Patients aged ≥12 years with newly diagnosed, melanoma were randomly assigned (1:1) 200 mg every weeks (2 mg/kg up pediatric patients) placebo. To establish a predictive model for prognosis of cutaneous melanoma using machine learning algorithms in large sample data. A retrospective analysis patients diagnosed with the SEER database from 2010 to 2015 was performed 12 different algorithms, total 97 algorithm combinations, screen variables associated and build models. 24,457 cases were collected this study, 8,441 finally included. Among them, 5908 training set 2533 test set. The results study show that StepCox[both] + RSF is best model. variable features screened by Sex, Age, Marital, T stage, N Ulcer, Site, Histologic, Surgery, Chemotherapy, Bone metastasis, Liver metastasis Lung metastasis. We have developed good accuracy combination database.
