The reduced form of nicotinamide adenine dinucleotide phosphate (NADPH) protects against redox stress by providing reducing equivalents to antioxidants such as glutathione and thioredoxin. NADPH levels decline with aging in several tissues, but whether this is a major driving force for the process has not been well established. Global or neural overexpression cytoplasmic enzymes that synthesize have shown extend lifespan model organisms Drosophila suggesting positive relationship between longevity. Evening-oriented sleep timing preferences have been associated with risk of diabetes, cardiovascular diseases, obesity, psychiatric disorders, and increased mortality. This research aims to explore the relationship between diurnal (chronotype), daily habits, metabolic health, mortality, using longitudinal data from The University Manchester Longitudinal Study Cognition in Normal Healthy Old Age (6375 participants at inception, recruited North England) a long follow-up period (up 35.5 years). Abstract Aging leads to changes in circadian rhythms, including decreased amplitude or robustness, altered synchrony with the environment, and reduced coordination of rhythms within body. These rhythm alterations are more pronounced age‐associated neurodegenerative disorders such as Alzheimer's disease ( AD ), which they often precede onset other symptoms by many years. As well their early onset, findings that fragmentation daily rest‐activity non‐demented older subjects is associated earlier cognitive decline, increased risk incident , preclinical neuropathology, suggest disruption may contribute development progression neuropathological occurring . Abstract The relationships between older age and sleep efficiency have traditionally been assessed using cross‐sectional studies that ignore changes within individuals as they age. Abstract Background CREB-dependent transcription necessary for long-term memory is driven by interactions with CREB-binding protein (CBP), a multi-domain that binds numerous factors potentially affecting expression of thousands genes. Identifying specific domain functions proteins essential to understand processes such as cognitive function and circadian clocks. We investigated the CBP KIX in hippocampal gene using KIX/KIX mice mutations prevent phospho-CREB (Ser133) binding. Results found were impaired memory, but not learning acquisition or short-term Morris water maze. Using an unbiased analysis dorsal hippocampus after training maze contextual fear conditioning, we discovered dysregulation CREB, CLOCK, BMAL1 target genes downregulation mice. Given our finding was important genes, profiled activity phase resetting exhibited delayed peaks light offset longer free-running periods constant dark. Interestingly, displayed delays advances response photic stimulation comparable wildtype littermates. Thus, this work delineates site-specific regulation clock protein. Conclusions These studies provide insight into significance defining targets transcriptional co-activation role vivo on rhythms. Graphical abstract
