(1) Background: It was found that the melanin of black-bone chicken has various effects such as scavenging DPPH free radicals and anti-oxidation, synthesis is affected by factors including hormones. In addition, several studies have melatonin affects melanoma cell melanin, which not been reported in primary melanocytes; so, relevant were conducted. (2) Methods: this study, melanocytes isolated characterized, then treated with different concentrations to investigate on terms synthesis-related genes, content, tyrosinase activity, combined RNA seq detect change gene expression level after treatment. (3) Results: We characterized melanocytes, indirect immunofluorescence assay results showed positive melanocyte marker genes. RT-qPCR decreased reduced content activity group. A total 1703 differentially expressed genes screened RNA-seq, KEGG results, signaling pathway associated synthesis, mTOR enriched. (4) Conclusions: Melatonin could decrease melanocytes.
