Osteosarcoma (OS) is the most prevalent secondary sarcoma associated with retinoblastoma (RB). However, molecular mechanisms driving interactions between these two diseases remain incompletely understood. This study aims to explore transcriptomic commonalities and pathways shared by RB OS, identify biomarkers that predict OS prognosis effectively. RNA sequences patient information for were obtained from University of California Santa Cruz (UCSC) Xena Gene Expression Omnibus databases. When first identified, a common gene expression profile was discovered. Weighted Co-expression Network Analysis (WGCNA) revealed co-expression networks after immunotyping patients. To evaluate genes univariate multivariate Cox regression analysis then carried out. Three machine learning methods used pick key genes, risk models created verified. Next, medications target independent prognostic found using Cellminer database. The comparison differential 1216 primarily linked activation proliferation immune cells. Ovarian cancer (OC), the gynecologic malignancy with poorest prognosis, is driven by metabolic reprogramming and dysregulated programmed cell death (PCD). However, their interplay prognostic significance remain inadequately understood. Transcriptomic data from OC patients healthy controls (TCGA GTEx) were analyzed to identify differentially expressed genes (DEGs) intersecting metabolism-related (MRGs) PCD-related (PCDRGs). Prognostic determined using univariate Cox regression, LASSO, multivariate stepwise analyses. Consensus clustering revealed enrichment differences, while a risk model nomogram developed for outcome prediction. Associations between genes, immune microenvironment, drug sensitivity also assessed. A total of 166 candidate identified, PLA2G2D, LPCAT3, ARG1, PLA2G4A, EXOSC3 emerging as significant markers. The demonstrated marked survival showed robust calibration Differential infiltration was observed groups. Additionally, Sinularin Fulvestrant exhibited variable sensitivity, validated through molecular docking models. Metabolism-related PCD identified pivotal markers in OC, providing critical insights evaluation targeted therapy development.

Abstract The role of NETosis and its related molecules remains unclear in gastric cancer. data used this study was directly downloaded from the Cancer Genome Atlas (TCGA) database. All analysis plots are completed R software using diverse packages. In our study, we collected list NETosis‐related genes previous publications. Based on expression profile cancer patients TCGA database, identified significantly correlated with survival. Then, CLEC6A, BST1 TLR7 were through LASSO regression multivariate Cox for prognosis model construction. This showed great predictive efficiency both training validation cohorts. We noticed that high‐risk might have a worse survival performance. Next, explored biological enrichment difference between high‐ low‐risk found many carcinogenic pathways upregulated patients. Meanwhile, investigated genomic instability, mutation burden immune microenvironment Moreover, more sensitive to immunotherapy (85.95% vs. 56.22%). High‐risk some small compounds like camptothecin_1003, cisplatin_1005, cytarabine_1006, nutlin‐3a (−)_1047, gemcitabine_1190, WZ4003_1614, selumetinib_1736 mitoxantrone_1810. summary, comprehensively cancer, which can provide direction relevant studies.

Background Neutrophil extracellular traps (NETs) have long been consistently considered an innate immune defense against foreign pathogens, but this oversimplified view has decelerated the progression of perceiving NET biology in chronic diseases. It is now increasingly accepted that NETs are not exclusive to anti-infection responses, also central players with a double-edged sword role cancer progression. gradually emerged as tumor diagnostic, predictive, and prognostic biomarkers, strenuous endeavors devoted tapping their potential new therapeutic targets. Correspondingly, boom studies on tumors recent years achieved series scientific outputs, which opens up perspective for sophisticated landscapes microenvironment.
