Corticotropin-releasing factor type-1 (CRF 1 ) receptors are critical to stress responses because they allow neurons respond CRF released in response stress. Our understanding of the role -expressing CRF-mediated behaviors has been largely limited mouse experiments due lack genetic tools available selectively visualize and manipulate + cells rats. Here, we describe generation validation a transgenic -Cre- td Tomato rat. We report that Crhr1 Cre mRNA expression highly colocalized both central amygdala (CeA), composed mostly GABAergic neurons, basolateral (BLA), glutamatergic neurons. In CeA, membrane properties, inhibitory synaptic transmission, bath application similar those previously reported GFP CRFR1-GFP mice. show stimulatory DREADD can be targeted CeA via virally delivered Cre-dependent transgenes, transfected Cre/ activated by clozapine-n-oxide vitro vivo, activation these vivo increases anxiety-like nocifensive behaviors. Outside amygdala, Cre- is expressed several brain areas across brain, pattern known cells. Given accuracy -Cre rat, modern techniques used investigate anatomy, physiology, behavioral function now performed assays require use rats as model organism.

Stress is a fact of life, affecting organisms from the smallest invertebrates to humans. Mediating stress system ancient neuropeptide, corticotropin releasing factor (CRF), which works as neuromodulator alter brain systems and homeostatic responses stress. In humans, many stress-related psychiatric illnesses are linked dysregulation monoamine systems, have cross-talk with CRF-enriched circuits. this review, we focus on CRF midbrain dopamine (DA) system, particularly it relates nonhuman primate. While tremendous knowledge CRF-DA mechanisms has been gleaned rodent models, treatments for diseases elusive, raising question whether higher animal models might be required. Subtle shifts in peptide or receptor localization, expansion complexity DA neuron populations, may hold some keys understanding long-standing effects Our laboratory especially interested laying out neural architecture interface primate, close anatomic model human. Using starting point, describe aspects complex that inform our interactions, results been, those still need be, translated primate models.
