Disorders of consciousness (DoC), including the unresponsive wakefulness syndrome (UWS) and minimally conscious state (MCS), have limited treatment options. Recent research suggests that psychedelic drugs, known for their complexity-enhancing properties, could be promising treatments DoC. This study uses whole-brain computational models to explore this potential. We created individualised DoC patients, optimised with empirical fMRI diffusion-weighted imaging (DWI) data, simulated administration LSD psilocybin. used an in-silico perturbation protocol distinguish between different states consciousness, DoC, anaesthesia, state, assess dynamical stability brains patients pre- post-psychedelic simulation. Our findings indicate psilocybin shift patients' closer criticality, a greater effect in MCS patients. In UWS response correlates structural connectivity, while it aligns baseline functional connectivity. virtual clinical trial lays foundation using psychedelics highlights future role modelling drug discovery personalised medicine.

Abstract A central challenge of neuroscience is to elucidate how brain function supports consciousness. Abstract Study Objectives This study aimed to investigate the alterations in resting-state electroencephalography (EEG) global brain connectivity (GBC) patients with chronic insomnia disorder (CID) and explore correlation between macroscale connectomic variances microscale neurotransmitter distributions.
