SUMMARY Advances in modern medical therapies for many previously intractable human diseases have improved patient outcomes. However, successful disease treatment outcomes are often prevented due to invasive fungal infections caused by the environmental mold Aspergillus fumigatus . As contemporary antifungal not experienced same robust advances as other therapies, defining mechanisms of A. initiation and progression remains a critical research priority. To this end, World Health Organization recently identified priority pathogen Centers Disease Control has highlighted emergence triazole-resistant isolates. The expansion diversity host populations susceptible aspergillosis complex dynamic genotypic phenotypic call reinvigorated assessment pathobiological drug-susceptibility mechanisms. Here, we summarize recent advancements field discuss challenges our understanding heterogeneity its pathogenesis diverse populations.

The environmental use of azole fungicides has led to selective sweeps across multiple loci in the Aspergillus fumigatus genome causing rapid global expansion a genetically distinct cluster resistant genotypes. Isolates within this are also more likely be agricultural antifungals with unrelated modes action. Here we show that is not only multi-azole but increased propensity develop resistance next generation because variants DNA mismatch repair system. A variant msh6-G233A found almost exclusively isolates harbouring canonical cyp51A allelic TR34/L98H. Naturally occurring msh6 display up 5-times higher rate mutation, leading an likelihood evolving other antifungals. Furthermore, unlike hypermutator strains, G233A conveys no measurable fitness cost and become globally distributed. Our findings further suggest next-generation emerge organisms already due close linkage between TR34/L98H msh6-G233A, posing major problem prospect dual novel clinical settings.

Azole resistance in Aspergillus fumigatus poses a significant clinical challenge globally. In this study, we analyzed 307 A. isolates from General Hospital of Ningxia Medical University China, collected between July 2023 and 2024, to explore their susceptibility profiles, genotypic characteristics biological traits. The overall frequency azole among province was found be 1.20% (7 azole-resistant fumigatus, designated AF1-AF7), each exhibiting distinct phenotypes terms resistance, spore viability, resilience environmental stress. Among these strains, the cyp51A mutations predominantly included TR43/L98H, while hmg1 were primarily S212P/Y564H. Additionally, novel discovered cyp51B, specifically t-215c point mutation base deletion (gatgccta) - 213 206 region. AF3 strain demonstrated highest activity anti-SDS efficacy. contrast, AF1 AF2 strains resistant three azoles also exhibited Menadione, similar AF7. Notably, six out seven displayed NaCl KCl, indicating strong tolerance saline conditions. These findings suggest that possess varying degrees stressors, implying they may adapt surroundings through different evolutionary pathways, which could complicate treatment strategies. Effective surveillance control strategies are essential widespread prevalence reduce risk infection patients.

ABSTRACT This review covers, for the first time, all methods based on use of Aspergillus strains as biocontrol agents management plant diseases caused by fungi and oomycetes. Atoxigenic have been screened in a variety hosts, such peanuts, maize kernels, legumes, during preharvest postharvest stages. These against wide range pathogens, Fusarium , Phytophthora Pythium species, suggesting broad applicability spectrum. The highest efficacies were generally observed when using non-toxigenic mycotoxin-producing strains. modes action included synthesis antifungal metabolites, kojic acid volatile organic compounds (VOCs), secretion hydrolytic enzymes, competition space nutrients, induction disease resistance. degraded Sclerotinia sclerotiorum sclerotia, showing high control efficacy this pathogen. Collectively, although two commercialized aflatoxin degradation, new application is emerging needs to be optimized.
