Shrimp aquaculture contributes significantly to global economic growth, and the whiteleg shrimp, Penaeus vannamei , is a leading species in this industry. However, Vibrio parahaemolyticus infection poses major challenge ensuring success of P . aquaculture. Despite its significance industry, biological knowledge pathogenesis remains unclear. Hence, study was conducted identify interaction sites binding affinity between several immune-related proteins with V associated virulence factors. Potential host pathogen were identified using molecular docking dynamics (MD) simulation. The vannamei-V protein-protein Complex 1 (Ferritin-HrpE/YscL family type III secretion apparatus protein), 2 (Protein kinase domain-containing protein-Chemotaxis CheY 3 (GPCR-Chemotaxis protein) found interact -4319.76, -5271.39, -4725.57 docked score formation intermolecular bonds at interacting residues. scores 1, 2, validated MD simulation analysis, which revealed these complexes greatly contribute interactions proteins, free energies -22.50 kJ/mol, -30.20 -26.27 respectively. This finding illustrates capability computational approaches search for pathogen, could increase spp. on shrimps, then can be used assist development effective treatment.

Quorum sensing (QS) is a process of cell-to-cell communication that bacteria use to synchronize collective behaviors. QS relies on the production, release, and group-wide detection extracellular signaling molecules called autoinducers. Vibrios two systems: LuxO-OpaR circuit VqmA-VqmR circuit. Both circuits control group behaviors including biofilm formation surface motility. The Vibrio parahaemolyticus temperate phage φVP882 encodes VqmA homolog (called VqmAφ). When VqmAφ produced by lysogens, it binds host-produced autoinducer DPO launches lytic cascade. This activity times induction lysis with high host cell density presumably promotes maximal transmission new cells. Here, we explore whether, in addition from lysogeny, controls initial establishment lysogeny naïve Using mutagenesis, infection assays, phenotypic analyses, show connects its lysis-lysogeny decision both whether resides liquid or surface. Host cells low-cell-density state primarily undergo lysogenic conversion. regulator LuxO~P conversion planktonic By contrast, ScrABC surface-sensing system regulates surface-associated abundance second messenger molecule cyclic diguanylate, which turn, modulates scrABC operon only expressed when repressor, OpaR, absent. Thus, at low density, QS-dependent derepression drives These results demonstrate integrates cues multiple sensory pathways into lifestyle making upon cell.

Vibrio parahaemolyticus must endure various challenging circumstances while being swallowed by phagocytes of the innate immune system. Moreover, bacteria should recognize and react to environmental signals quickly in host cells. Two-component system (TCS) is an important way for perceive external transmit them interior trigger associated regulatory mechanism. However, function V. TCS cells unclear. Here, expression patterns -infected THP-1 cell-derived macrophages at early stage were studied first time. Based on protein-protein interaction network analysis, we mined analyzed seven critical genes with excellent research value regulating macrophages, as shown below. VP1503, VP1502, VPA0021, VPA0182 could regulate ATP-binding-cassette (ABC) transport VP1735, uvrY, peuR might interact thermostable hemolysin proteins, DNA cleavage-related TonB-dependent siderophore enterobactin receptor, respectively, which may assist infected macrophages. Subsequently, potential escape pathways explored RNA-seq. The results showed that infect controlling apoptosis, actin cytoskeleton, cytokines. In addition, found (peuS/R) enhance toxicity contribute activation macrophage apoptosis. IMPORTANCE This study offer crucial new insights into pathogenicity without tdh trh genes. also provided a novel direction inquiry pathogenic mechanism suggested several key regulation interaction.
