Porcine Epidemic Diarrhea Virus (PEDV) and Transmissible Gastroenteritis (TGEV) pose significant threats to neonatal piglets, leading severe diarrhea potentially lethal consequences. Beyond enforcing stringent biosecurity protocols, effective safe vaccinations are crucial in mitigating the impact of these diseases. In this study, PEDV S1 (PS1) TGEV (TS1) antigens were initially chosen as candidates for development circRNA vaccines. Recognizing comparatively lower immunogenicity PS1 antigen contrast TS1 antigen, we strategically conjugated with pig fragment crystallizable (Fc) region form PS1F. Despite efforts, bivalent vaccine prepared using an equal amount circRNAPS1F circRNATS1 mixture still led a reduction antibody levels against PS1. Subsequent dosage optimization two vaccines resulted induction comparable specific antibodies T cell immunity. Tuberculosis (TB) is caused by infection with the bacterial pathogen Mycobacterium tuberculosis (M.tb) in respiratory tract. There was an estimated 10.6 million people newly diagnosed TB, and there were approximately 1.3 deaths TB 2022. Although global prevalence of has remained high for decades annual leading cause death attributed to infectious diseases, only one vaccine, Bacillus Calmette–Guérin (BCG), been approved so far prevent/attenuate disease. Marburg virus (MARV), a filovirus, was first identified in 1967 Marburg, Germany, and Belgrade, former Yugoslavia. Since then, MARV has caused sporadic outbreaks of human disease with high case fatality rates parts Africa, the largest outbreak occurring 2004/05 Angola. There are currently no approved vaccines or therapeutics targeting MARV, but several vaccine candidates have shown promise preclinical studies. We compared three platforms simultaneously by vaccinating hamsters either single dose an adenovirus-based (ChAdOx-1 MARV) vaccine, alphavirus replicon-based RNA (LION-MARV) recombinant vesicular stomatitis virus-based (VSV-MARV) all expressing glycoprotein as antigen. Lethal challenge hamster-adapted 4 weeks after vaccination resulted uniform protection VSV-MARV LION-MARV groups 83% ChAdOx-1 group. Assessment antigen-specific humoral response functionality revealed vaccine-platform-dependent differences, particularly Fc effector functions.

Immunity acquired by vaccination following infection, termed hybrid immunity, has been shown to confer enhanced protection against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) enhancing the breadth and potency of immune responses. Here, we assess Fc-mediated humoral profiles in immunity their association with age vaccine type. Participants are divided into three groups: infection only, (i.e., immunity).
