Plesiomonas shigelloides, a Gram-negative bacillus, is the only member of Enterobacteriaceae family able to produce polar and lateral flagella cause gastrointestinal extraintestinal illnesses in humans. The flagellar transcriptional hierarchy P. shigelloides currently unknown. In this study, we identified FlaK, FlaM, FliA, FliAL as four regulators responsible for regulation shigelloides. To determine transcription transcriptomes WT ΔflaK, ΔflaM, ΔfliA, ΔfliAL were carried out comparison study. Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) luminescence screening assays used validate RNA-seq results, Electrophoretic Mobility Shift Assay (EMSA) results revealed that FlaK can directly bind promoters fliK, fliE, flhA, cheY, while FlaM protein flgO, flgT, flgA. Meanwhile, also observed type VI secretion system (T6SS) II 2 (T2SS-2) genes downregulated transcriptome profiles, killing assay lower abilities compared WT, indicating there was cross-talk between bacterial system. Invasion showed less effective infecting Caco-2 cells than WT. Additionally, found loss causes differential expression some physiological metabolic Overall, study aims reveal controls gene well motility, virulence, activity, laying groundwork future research into shigelloides' coordinated survival natural environment mechanisms infect host.
