Identifying and mapping steroids in tissues can provide opportunities for biomarker discovery, the interrogation of disease progression, new therapeutics. Although separation coupled to mass spectrometry (MS) has emerged as a powerful tool studying steroids, imaging annotating steroid isomers remains challenging. Herein, we present method based on fragmentation silver-cationized tandem MS, which produces distinctive consistent patterns conferring confidence annotation at regioisomeric level without using prior derivatization, separation, or instrumental modification. In addition predicting structure with isomeric specificity, is simple, flexible, inexpensive, suggesting that wider community will easily adapt it. We demonstrate utility our approach by visualizing isomer distributions mouse brain tissue silver-doped pneumatically assisted nanospray desorption electrospray ionization imaging.
