Abstract Metabolic homeostasis is under strict regulation of humoral factors across various taxa. In particular, insulin and glucagon, referred to in Drosophila as insulin‐like peptides (DILPs) adipokinetic hormone (AKH), respectively, are key hormones that regulate metabolism most metazoa. While much known about the DILPs, mechanisms regulating AKH/glucagon production still poorly understood. this review, we describe DILPs AKH emphasize need for future studies decipher how energy governed . Carbohydrate metabolism is essential for cellular energy balance as well the biosynthesis of new building blocks. As animal nutrient intake displays temporal fluctuations and each cell type within possesses specific metabolic needs, elaborate regulatory systems are needed to coordinate carbohydrate in time space. regulated locally through gene networks signaling pathways, which receive inputs from sensors other such developmental signals. Superimposed on cell-intrinsic control, hormonal mediates intertissue information maintain organismal homeostasis. Misregulation causative many human diseases, diabetes cancer. Recent work Drosophila melanogaster has uncovered regulators introduced novel physiological roles previously known pathways. Moreover, genetically tractable models study metabolism-related diseases have provided insight into mechanisms pathogenesis. Due high degree conservation relevant vast possibilities analysis gene-nutrient interactions tissue-specific function, emerging an important model system research metabolism.

Physiology and metabolism are often sexually dimorphic, but the underlying mechanisms remain incompletely understood. Here, we use intestine of Drosophila melanogaster to investigate how gut-derived signals contribute sex differences in whole-body physiology. We find that carbohydrate handling is male-biased a specific portion intestine. In contrast known sexual dimorphisms invertebrates, intestinal extrinsically controlled by adjacent male gonad, which activates JAK-STAT signaling enterocytes within this portion. Sex reversal experiments establish roles for metabolic state controlling food intake sperm production through citrate. Our work uncovers gonad-gut axis coupling diet production, revealing communication across organs physiologically important. The instructive role citrate inter-organ might be significant more biological contexts than previously recognized.

Adapting to changes in food availability is a central challenge for survival. Glucose an important resource energy production, and therefore many organisms synthesize retain sugar storage molecules. In insects, glucose stored two different forms: the disaccharide trehalose branched polymer glycogen. Glycogen synthesized several tissues, including muscle fat body. Despite major role of body as center metabolism, importance its glycogen content remains unclear. Here, we show that metabolism regulated tissue-specific manner under starvation conditions fruit fly Drosophila. The mobilization larvae independent Adipokinetic hormone (Akh, glucagon homolog) but by tissue-autonomous manner. Fat plays crucial maintenance circulating sugars, trehalose, fasting conditions. These results demonstrate metabolic safeguard
