Candida albicans, a ubiquitous opportunistic fungal pathogen, plays pivotal role in human health and disease. As commensal organism, it normally resides harmlessly within the microbiota. However, under certain conditions, C. albicans can transition into pathogenic state, leading to various infections collectively known as candidiasis. With increasing prevalence of immunocompromised individuals widespread use invasive medical procedures, candidiasis has become significant public concern. The emergence drug-resistant strains further complicates treatment options, highlighting urgent need for alternative therapeutic strategies. Antifungal peptides (AFPs) have gained considerable attention potential candidates combating spp. infections. These naturally occurring possess broad-spectrum antimicrobial activity, including specific efficacy against albicans. AFPs exhibit several advantageous properties, such rapid killing kinetics, low propensity resistance development, diverse mechanisms action, making them promising alternatives conventional antifungal agents. In recent years, extensive research focused on discovering developing novel with improved selectivity species. Advances biotechnology synthetic peptide design enabled modification optimization natural peptides, enhancing their stability, bioavailability, potential. Nevertheless, challenges must be addressed before widely implemented clinical practice. include optimizing delivery methods, overcoming toxicity concerns, conducting comprehensive preclinical studies. The incidence of invasive fungal diseases (IFDs) is on the rise globally, particularly among immunocompromised patients, leading to significant morbidity and mortality. Current clinical antifungal agents, such as polyenes, azoles, echinocandins, face increasing resistance from pathogenic fungi. Therefore, there a pressing need for development novel drugs. Marine-derived secondary metabolites represent valuable resources that are characterized by varied chemical structures pharmacological activities. While numerous compounds exhibiting promising activity have been identified, comprehensive review elucidating their specific underlying mechanisms remains lacking. In this review, we compiled summary derived marine organisms, highlighting diverse action targeting various cellular components, including cell wall, membrane, mitochondria, chromosomes, drug efflux pumps, several biological processes, vesicular trafficking growth hyphae biofilms. The adverse outcomes of fungal infection in mammalian hosts depend on the complex interactions between host immune system and pathogen virulence-associated traits. main clinical problems arise when response is either too weak to effectively eliminate or overly aggressive, resulting tissue damage rather than protection. This article will highlight current knowledge regarding virulence attributions mechanisms involved dual-sided role immunopathogenesis thermally dimorphic fungus Talaromyces marneffei through lens framework (DRF) microbial pathogenesis model.
