This study employed multivariate analysis to investigate the effects of a 14-week multicomponent training program on sedentary adults and older populations, focusing improvements in physical fitness, health markers, anthropometric measures. The data included 376 participants aged 30 84 years who had been inactive for at least 3 months. Results showed that 71 above lower systolic blood pressure, height, body mass, elbow flexion extension, sitting standing up from chair, agility, 6-Minute Walk Test (6MWT), handgrip strength values compared younger age groups (p < 0.05). Initially, 6MWT was inversely correlated with measures positively tests, but after intervention, it remained only agility. Interestingly, extension strongly other fitness pre-intervention, these relationships weakened afterward. Agility became key intermediary variable analysis. Despite improvements, variables like mass waist circumference continued have strong relationship outcomes. Significant gains were observed 0.05), indicating aerobic strength. Multivariate hip circumference, chair stand test, strength, agility significantly influenced performance. Using analysis, we able highlight importance incorporating regular exercise into daily life promote active healthy aging. Furthermore, emerged as most significant intermediate intervention. insight suggests may be crucial overall health.

Exercise training emerges as a key strategy in lifestyle modification, capable of reducing the risk developing Alzheimer's disease (AD) due to factors such age, family history, genetics and low level education associated with AD. We aim analyze effect 14-week combined exercise (CT) on methylation genes AD non-alzheimer's women. CT sessions lasted 60 min, occurring three times week for 14 weeks. Forty non-Alzheimer's women aged 50 70 years (60.7 ± 4.1 years) mean height 1.6 0.1 m, weight 73.12 9.0 kg body mass index 29.69 3.5 kg/m2, underwent two physical assessments: pre post DNA assays utilized EPIC Infinium Methylation BeadChip from Illumina. observed that weeks led reductions systolic (p = 0.001) diastolic 0.017) blood pressure improved motor skills post-intervention. Among 25 linked AD, induced differentially methylated sites 12 genes, predominantly showing hypomethylated (negative β values). Interestingly, despite sites, some exhibited hypermethylated (positive values), ABCA7, BDNF, WWOX. A regimen was adequate induce differential CE-related healthy older women, alongside improvements pressure. In conclusion, this study suggest can be improve fitness individuals, especially able alterations related development It is important highlight should act protective factor adults.

Purpose: The acceleration of epigenetic age is a predictor mortality and contributes to the increase in chronic diseases. Adherence healthy lifestyle strategy reduce age. present study aimed determine whether eight weeks combined (aerobic strength) training (CT) can influence women between 50 70 years old differences sites methylated regions. Methods: Eighteen (AARLow: lower residual, n = 10; AARHigh: higher 8) participated exercise program (60 minutes, 3× week) for weeks. DNA was extracted from whole blood using salting out technique. methylation performed array technique (Illumina's Infinium Methylation BeadChip 850k). We used Age Calculator platform calculate biological Two-way ANOVA followed by FISHER LSD posthoc Applied, adopting p < .05. Results: After CT, there were no changes AARLow group (PRE: -2.3 ± 3.2 POST: 3.6). However, AARHigh significantly decreased 3.6 2.6 2.2 2.7) (group effect, .01; time .31; vs. .005). Introduction: The decrease in lean mass is directly related to the loss of independence, muscle strength, and worse quality life over years. Although genetic determinants were well recognized, recent literature has been uncovering new epigenetic factors affecting state muscular tissue. This study aimed verify differences DNA methylation profile among Brazilian postmenopausal women aged 50–70 years according evaluation. Methods: A cross-sectional comprised 40 After K-means cluster analysis participants divided into two groups, Lower Lean Mass group with 20 (61.1 ± 4.6 years) Higher (60.7 3.2 years). was measured by dual-energy X-ray emission densitometry (DEXA). participants' extracted using Salting Out technique subsequently, Illumina 850k EPIC Infinium Methylation BeadChip performed obtain data. Results: We obtained 1,913 differentially methylated sites ( p ≤ 0.005 β > 5% < −5%) a total 979 genes between groups 0.005; −5% 5%). In addition, PI3K-Akt pathway had greatest power significance an FDR × 10 –3 . Conclusion: Our results demonstrate differentiation specific different genes, which have essential functions body composition energy metabolism, supporting future studies that aim relate epigenetics.

Abstract Motivation DNA methylation plays a crucial role in biological processes, including imprinting, development, inflammation, and several disorders, such as cancer. Bisulfite sequencing (BS) is the gold standard for single-base resolution measuring methylation. This process involves treating genomic with sodium bisulfite, followed by polymerase chain reaction (PCR), converting unmethylated cytosines to thymines (C T) guanines adenines (G A). However, aligning reads obtained through next-generation (NGS) of converted challenging due high number mismatches caused these conversions. Various BS aligners aim simplify read alignment conventional modifying reference genome and/or reads. Methods include three-letter wild-card alignment, each its limitations. work introduces novel aligner, ARYANA-BS, which departs from considering base alterations within engine. Leveraging well-established patterns different contexts, ARYANA-BS generates five indexes reference, aligns all indexes, selects hit minimum penalty. To further enhance accuracy, an optional EM step has been developed, incorporating probability information decision-making index penalty read. The presented approach seeks improve accuracy accounting intricacies diverse contexts. Results Our experimental results, based on both simulated real data, demonstrate that achieves state-of-the-art while maintaining competitive speed memory usage.
