The aim of the present study was to evaluate protective effect gallic acid (GA) against cisplatin (CDDP)-induced ovarian toxicity, for first time. damage generated with CDDP (5 mg/kg) intraperitoneally (i.p.) administration in rats. GA (2.5 and 5 were administered i.p. 3 consecutive days. carried out main groups containing 6 rats each group: control, mg/kg), CDDP, + mg/kg). levels malondialdehyde (MDA), total oxidant status (TOS), antioxidant (TAS), oxidative stress index (OSI), catalase (CAT), 8-hydroxy-2'-deoxyguanosine (8-OHdG), caspase-3 tumor necrosis factor-alpha (TNF-α) determined. Hematoxylin eosin staining method employed histopathological examination. In group, it is determined that statistically significant decreasing TAS CAT, increasing MDA, TOS, OSI, 8-OHdG, TNF-α (p < 0.05) compared control group. administrations significantly restored this 0.05). Although vascular congestion, edema, hemorrhage, follicular degeneration leukocyte infiltration higher group than these damages conclusion, showed prevented CDDP-induced its antioxidant, anti-apoptotic anti-inflammatory activities. The global prevalence of type 2 diabetes mellitus (T2DM) and Alzheimer’s disease (AD) is rapidly increasing, revealing a strong association between these two diseases. Currently, there are no curative medication available for the comorbidity T2DM AD. Ceramides structural components cell membrane lipids act as signal molecules regulating homeostasis. Their synthesis degradation play crucial roles in maintaining metabolic balance vivo , serving important mediators development neurodegenerative disorders. Abnormal ceramide metabolism disrupts intracellular signaling, induces oxidative stress, activates inflammatory factors, impacts glucose lipid homeostasis metabolism-related tissues like liver, skeletal muscle, adipose tissue, driving occurrence progression T2DM. connection changes levels brain, amyloid β accumulation, tau hyper-phosphorylation evident. The therapeutic value of cisplatin (CDDP) as an anticancer drug is limited by its ovo-otoxicity. effect natural phenolic acids in the prevention many diseases related to oxidative stress has been reported. Here, ability p-coumaric (pCA) acid, a member acids, protect rat ovary tissue against CDDP-induced was investigated. The study carried out five main groups containing six rats each group: control, pCA (4 mg/kg), CDDP, CDDP plus (2 and mg/kg). levels ovarian malondialdehyde (MDA), total oxidant status (TOS), antioxidant (TAS), index (OSI), catalase (CAT), 8-hydroxy-2'-deoxyguanosine (8-OHdG), caspase-3, tumor necrosis factor-alpha (TNF-α) were determined. Hematoxylin eosin staining method employed for histopathological examination. In group, it determined that statistically significant decreasing TAS CAT, increasing MDA, TOS, OSI, 8-OHdG, TNF-α compared with control group (p < 0.05). administration significantly restored this damage dose-dependent manner Although vascular congestion, edema, hemorrhage, follicular degeneration, leukocyte infiltration higher than administrations these damages 0.05).The data presented here indicate protects tissues stress, inflammation, apoptosis. It may be worthy consider usefulness adjuvant therapy cancer management.

Growing evidence suggests that a high fat diet (HFD) induces oxidative stress on the central nervous system (CNS), which predisposes to mood disorders and neuroinflammation. In this study we postulated in addition improving mood, antidepressant therapy would reverse inflammatory changes brain of rats exposed HFD. To test our hypothesis, measured effect agomelatine (AGO) anxiety- depressive-like behaviors, as well CNS markers inflammation rendered obese. Agomelatine is an agonist melatonin receptors MT1 MT2 antagonist serotonin 5HT2B 5HT2C. A subset was also treated with lipopolysaccharides (LPS) determine how additional neuroinflammation alters behavior affects response antidepressant. Specifically, were subjected 14-week HFD, during time evaluated twice, first at 10th week prior LPS and/or agomelatine, then 14th after bi-weekly exposure (250 μg/kg) daily treatment (40 mg/kg). Immediately second behavioral testing proinflammatory cytokines tumor necrosis factor alpha (TNF-α), interleukin 6 (IL-6) 1 beta (IL-1β), thiobarbituric acid reactive substances (TABRS), catalase (CAT) glutathione peroxidase (GPx), growth BDNF, apoptosis marker caspase-3. Our results show HFD induced anxiety-like open field (OFT) week, followed by forced swim (FST) week. prefrontal hippocampal cortices noted overproduction TNF-α, IL-6, IL-1β, TABRS, together increase caspase-3 activity. We observed decrease reduced CAT GPx activity same areas. Treatment reversed signs anxiety depression, decreased (TNF-α, IL-6 IL-1β), restored BDNF levels antioxidant enzymes GPx. findings suggest anxiolytic/antidepressant obese could result from reversal brought about their diet.
