Background: The causal relationships between amyotrophic lateral sclerosis (ALS), Parkinson disease and different intensities of physical activity (PA) are still inconclusive. To evaluate the impact PA on ALS (PD), this study integrates evidence from Mendelian randomization (MR) using a meta-analysis approach. Methods: MR analyses genetically predicted levels (compose self-reported moderate-to-vigorous [MVPA], vigorous [VPA], strenuous sports or other exercises [SSOE]) regarding PD published up to July 27, 2024, were obtained PubMed, Scopus, Web Science, Embase. De novo studies analyzed utilizing publicly accessible datasets genome-wide association then meta-analyses performed pool results. Results: Meta-analyses results 12 de 2 indicated that genetic MVPA (odds ratio [OR]: 1.22, 95% confidence interval [CI]: 1.08–1.38), VPA (OR: 1.32, CI: 1.08–1.60), SSOE 1.35, 1.07–1.70) related raised risk ALS, but not causally with PD. Conclusion: Our findings showed no MVPA, VPA, SSOE, PD, while associated increased risk, highlighting need for targeted recommendations management.
