Growing elderly populations are an increasingly serious health and socio-economic concern for modern societies. Science has made tremendous progress in the understanding of aging itself which somehow helped medicine to extend life expectancies. Inflammaging refers to a chronic, systemic, low-grade inflammation, driven by immune (mainly macrophages) and non-immune cells stimulated endogenous/self, misplaced or altered molecules, belonging physiological aging. This age-related inflammatory status is characterized increased inflammation decreased macroautophagy/autophagy (a degradation process that removes unnecessary dysfunctional cell components). predisposes diseases, including obesity, type-2 diabetes, cancer, cardiovascular neurodegenerative disorders, as well vulnerability infectious diseases vaccine failure, representing thus major target for anti-aging strategies. Phenolic compounds—found in extra-virgin olive oil (EVOO)—are known their beneficial effect on longevity. Among them, hydroxytyrosol (HTyr) appears greatly contribute healthy aging its documented potent antioxidant activity. In addition, HTyr can modulate autophagy, possibly counteracting reducing inflammaging. this review, we reference the literature pure modulatory agent of order highlight possible interference with HTyr-mediated activity might delay development progression related

High dietary intake of saturated fatty acids is a suspected risk factor for neurodegenerative diseases, including Alzheimer’s disease (AD). To decipher the causal link behind these associations, high-fat diets (HFD) have been repeatedly investigated in animal models. Preclinical studies allow full control over composition, avoiding ethical concerns clinical trials. The goal present article to provide narrative review reports on HFD models AD. Eligibility criteria included mouse AD fed defined as > 35% fat/weight and western containing 1% cholesterol or 15% sugar. MEDLINE Embase databases were searched from 1946 August 2022, 32 preclinical review. HFD-induced obesity metabolic disturbances such insulin resistance glucose intolerance replicated most studies, but with methodological variability. Most found an aggravating effect brain Aβ pathology, whereas tau pathology has much less studied, results are more equivocal. While show impairment cognitive behavior, confounding factors may blur their interpretation. In summary, despite conflicting results, exposing rodents highly enriched fat induces not only defects, also often accompanied by aggravated neuropathological markers, notably burden. Although there important variations between methods, particularly lack diet characterization, collectively suggest that excessive should be avoided order lower incidence

Aging is characterized by a chronic low-grade sterile inflammation dubbed as inflammaging, which in part originates from accumulating cellular debris. These, acting danger signals with many intrinsic factors such cytokines, are sensed network of pattern recognition receptors and other cognate receptors, leading to the activation inflammasomes. Due inflammasome activity-dependent increase levels pro-inflammatory interleukins (IL-1β, IL-18), initiated, resulting tissue injury various organs, brain spinal cord included. Similarly, age-related diseases central nervous system (CNS), prominent moment, cells neurovascular unit occupy significant position. In this review, we discuss inflammatory changes normal aging summarize current knowledge on role inflammasomes contributing mechanisms common CNS diseases, namely Alzheimer’s disease, Parkinson’s amyotrophic lateral sclerosis stroke, all occur more frequently aging.
