ABSTRACT
Congenital
microcoria
(MCOR)
is
a
rare
hereditary
developmental
defect
of
the
iris
dilator
muscle,
frequently
associated
with
high
axial
myopia
and
intraocular
pressure
(IOP)
glaucoma.
The
condition
caused
by
submicroscopic
rearrangements
chromosome
13q32.1.
However,
mechanisms
underlying
failure
development
origin
features
remain
elusive.
Here,
we
present
3D
architecture
model
13q32.1
region,
demonstrating
that
MCOR-related
deletions
consistently
disrupt
boundary
between
two
Topologically
Associating
Domains
(TADs).
Deleting
critical
MCOR-causing
region
in
mice
reveals
ectopic
Sox21
expression
precisely
aligning
Dct
,
each
located
one
neighbor
TADs.
This
observation
consistent
TADs’
alteration
adoption
regulatory
elements
promoter.
Additionally,
identify
Tgfb2
as
target
gene
SOX21
show
TGFB2
accumulation
aqueous
humor
MCOR-affected
subject.
Accumulation
recognized
for
its
role
glaucoma
potential
impact
on
myopia.
Our
results
highlight
importance
SOX21-TGFB2
signaling
control
eye
growth
IOP.
Insights
from
MCOR
studies
may
provide
therapeutic
avenues
this

ABSTRACT
Objective
To
investigate

cytokines

aqueous
humor

Abstract
Purpose
Endoplasmic
reticulum
(ER)
stress,
mitochondrial
dysfunction,
mitophagy/autophagy
are
known
to
contribute
independently
corneal
endothelial
(CE)
apoptosis
in
Fuchs’
dystrophy
(FECD).
However,
the
role
of
a
well-studied
specific
ER
stress
pathway
(PERK-ATF4-CHOP)
regulating
mitophagy/autophagy,

unknown.
