Abstract INTRODUCTION Although sleep disturbances are widely recognized as risk factors for cognitive decline and Alzheimer's disease (AD), their influence on AD biomarkers remains unclear. This study aimed to clarify whether quality or duration affect amyloid beta (Aβ) tau levels in plasma, cerebrospinal fluid (CSF), positron emission tomography (PET) non‐demented populations. METHODS PubMed, Web of Science, Embase were systematically searched up February 2025. RESULTS In total, 30 studies included comprising 14,997 subjects. Individuals with poor exhibited greater PET Aβ burden higher Aβ42 plasma than those good quality. Shorter was associated PET. However, no association between either found. DISCUSSION Sleep may be a modifiable marker early management by modulating levels. Highlights lPoor shorter significantly detected Poor also elevated plasma. lNo significant associations found fluid, lInterventions targeting could serve viable low‐cost prevention strategy disease.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by cognitive decline, beta-amyloid plaques, and tau tangles. Growing evidence suggests strong link between sleep disturbances AD progression, with disrupted exacerbating progression through increased accumulation. This relationship indicates that improving quality could slow mitigate its effects on the brain. We investigated whether vestibular stimulation (rocking) pathology in 3xTg mice (n = 58, males). Starting early adulthood (p60), underwent 12-h daily rocking during light period for four months. Rocking non-rapid eye movement (NREM) initially, although habituation reduced this effect over time. Despite habituation, slowed motor decline levels cerebral cortex hippocampus. However, remained unaffected. In conclusion, our findings highlight potential of non-pharmacological methods to enhance NREM modify trajectory models.

Abstract Alzheimer’s disease (AD) is a progressive neurodegenerative disorder associated with cognitive decline and characterized by beta-amyloid plaque tau tangle pathology. Recent research indicates bidirectional relationship between AD pathology sleep disturbances, disrupted exacerbating progression through increased accumulation. This strongly that improving may exert direct protective effect on preventing the accumulation spreading of pathology, possibly slow decline. Here we investigated effects enhancing via vestibular stimulation (rocking) in 3xTg mouse model. Over four-month period starting early adulthood (p60), monitored patterns, motor function, memory, Twelve-hour rocking during light significantly non-rapid eye movement (NREM) duration mice, although this diminished over time due to habituation. Despite this, attenuated function reduced levels cerebral cortex treated mice. No noticeable changes were observed following enhancement. In conclusion, our findings highlight potential non-pharmacological methods enhance NREM modify trajectory models, emphasizing critical role neuroprotection.
