A large fraction of the human genome is transcribed in RNA molecules that do not encode for proteins but have a crucial role regulating almost every level gene expression and, thus, define specific phenotype each cell. These non-coding RNAs include well-characterized microRNAs and thousands less-defined longer transcripts, named long RNAs. Both types markedly affect onset progression numerous pathologies, ranging from cancer to vascular neuro-degenerative diseases. In recent years, substantial effort has been made design drugs targeting ncRNAs, promising advancements produced micro-RNA mimics inhibitors. Despite being one of the primary and most effective treatments for advanced stages lung cancer, chemotherapy drugs like carboplatin have limitations due to their adverse side effects development drug resistance in cancer cells. However, recent studies shown promising results using small interfering RNAs (siRNAs) as a therapeutic agent treatment. Hence, this study aimed investigate potential combining siRNA-DLGAP1-AS2 with human cell lines. The viability cells was assessed MTT assay, apoptosis induction examined through Annexin V/Pi staining. Additionally, effect combination on cycle arrest colony formation studied. Furthermore, expression Bax, Bcl-2, MMP-2, MMP-9, GCLC, CD44 evaluated. Our functional analysis revealed that inhibiting DLGAP1-AS2 increased sensitivity carboplatin. Moreover, our demonstrated inhibition transfection treatment had tumor-suppressive function, progression proliferation A549 Therefore, it can be concluded targeting specific siRNA holds promise valuable approach cancer.

MicroRNAs (miRNAs) have emerged as crucial regulators in digestive pathologies, including inflammatory bowel disease (miR-31, miR-155, and miR-21), colorectal cancer (miR-21, miR-598, miR-494), non-alcoholic fatty liver miR-192, miR-122). Their capacity to modulate gene expression at the post-transcriptional level makes them highly promising candidates for biomarkers therapeutic interventions. However, despite considerable progress, their clinical application remains challenging. Research has shown that miRNA is dynamic, varying across patients, stages, different intestinal regions. dual function both oncogenes tumor suppressors further complicates use, targeting miRNAs may yield unpredictable effects. Additionally, while miRNA-based therapies hold great potential, significant hurdles persist, off-target effects, immune activation, inefficiencies delivery methods. The intricate interplay between gut microbiota adds another layer of complexity, influencing mechanisms treatment responses. This review examined role emphasizing diagnostic potential. While they offer new avenues management, unresolved challenges underscore need research refine application.
