Modification of the novel N6-methyladenine (m6A) DNA implicates this epigenetic mark in human malignant disease, but its role atherosclerosis (AS) is largely unknown. Here, we found that leukocyte level m6A not 5mC modification was decreased with increasing carotid plaque size and thickness 207 AS patients as compared 142 sex- age-matched controls. Serum low-density lipoprotein (LDL) levels were associated progression thickness. Both LDL also independently negatively related to level. Reduced further confirmed leukocytes endothelium western diet-induced mice oxidized-LDL (ox-LDL)-treated monocyte cells. Decreased closely upregulation AlkB homolog 1 (ALKBH1), demethylase m6A. Silencing ALKBH1 or hypoxia-inducible factor 1α (HIF1α) could rescue ox-LDL-increased MIAT, a hypoxia-response gene. Mechanically, ox-LDL induced HIF1α for transfer into nucleus. Nuclear bound ALKBH1-demethylated MIAT promoter transcriptionally upregulated expression. Therefore, elevated reduced level, which sensitive biomarker progression.

Abstract Resistance of chemotherapy is one causes recurrence and poor prognosis in patients with colorectal cancer (CRC). The role differentially expressed long non-coding RNA (lncRNA) 5-fluorouracil (5-Fu) resistance has not been fully elucidated. Here we observed that lncRNA NEAT1 was associated 5-Fu CRC. Our Functional studies showed promoted cells. In addition, A-TAC sequencing chromatin immunoprecipitation (ChIP) affected remodeling, increased the acetylation levels histones, their enrichment at promoters ALDH1 c-Myc, expression c-Myc. Taken together, our study suggested stemness by remodeling chromatin. finding provides a novel may provide new strategy for treatment CRC resistance.

However, accurate role miR-361 CC-associated EMT mechanisms underlying its function CC remains largely unknown. The functional roles cells were explored by series cell assays. Luciferase reporter assays used demonstrate potential interaction between miR-361, HSP90, long non-coding RNA (lncRNA) NEAT1. We detected reduction tissues compared with normal tissues, overexpression inhibited invasion phenotypes directly targeting activator HSP90. Additionally, we significantly higher levels HSP90 high predicted poorer prognosis. further identified NEAT1 as upregulated lncRNA was associated worse survival patients. repressed played an oncogenic sphere formation. Conclusions: These results demonstrated that targets inhibit features, functions suppresses induces formation cells, thus providing critical insights into molecular pathways operating this malignancy.
