Circular RNA (circRNA) is a highly abundant type of single-stranded non-coding RNA. Novel research has discovered many roles circRNA in colorectal cancer (CRC) including proliferation, metastasis and apoptosis. Furthermore, circRNAs also play role the development drug resistance have unique associations with tumour size, staging overall survival CRC that lend potential to serve as diagnostic prognostic biomarkers. Among cancers worldwide, ranks second mortality third incidence. In order better understanding influence on progression, this review summarizes specific evaluates their value therapeutic targets biomarkers for CRC. We aim provide insight therapy clinical decision-making.

Abstract Previous studies have demonstrated the highly specific expression of circular RNAs (circRNAs) in different tissues and organisms, but cellular architecture circRNA has never been fully characterized. Here, we present a collection 171 full-length single-cell RNA-seq datasets to explore landscape circRNAs human mouse tissues. Through large-scale integrative analysis, identify total 139,643 214,747 these scRNA-seq libraries. We validate detected with integration 11 bulk based resources, where 216,602 high-confidence are uniquely cohort. reveal cell-type-specific pattern brain samples, developing embryos, breast tumors. expressed cell types their performance tumor-infiltrating immune composition deconvolution. This study expands our knowledge level provides useful resource for exploring at this unprecedented resolution.

Circular RNA (circRNA) is closely associated with human diseases. Accordingly, identifying the associations between diseases and circRNA can help in disease prevention, diagnosis treatment. Traditional methods are time consuming laborious. Meanwhile, computational models effectively predict potential circRNA-disease (CDAs), but restricted by limited data, resulting data high dimension imbalance. In this study, we propose a model based on automatically selected meta-path contrastive learning, called MPCLCDA model. First, constructs new heterogeneous network similarity, similarity known association, via obtains low-dimensional fusion features of nodes graph convolutional networks. Then, learning used to optimize further, obtain node that make distinction positive negative samples more evident. Finally, scores predicted through multilayer perceptron. The proposed method compared advanced four datasets. average area under receiver operating characteristic curve, precision-recall curve F1 score 5-fold cross-validation reached 0.9752, 0.9831 0.9745, respectively. Simultaneously, case studies further prove predictive ability application value method.

Cardiovascular disease is one of the leading causes mortality worldwide. Recent studies have shown that circular RNAs (circRNAs) emerged as important players in prevention and treatment cardiovascular diseases. circRNAs are a class endogenous noncoding generated by back-splicing involved many pathophysiological processes. In this review, we outline current research progress on regulatory roles Further, new technologies methods available for identifying, validating, synthesizing, analyzing circRNAs, well their applications therapeutics, highlighted here. Moreover, summarize increasing insights into potential use circulating diagnostic prognostic biomarkers. Finally, discuss prospects challenges circRNA therapeutic therapy, with particular focus developing synthesis engineering delivery systems.
