A number of studies have demonstrated that circular RNA (circRNA) plays a critical role in tumorigenesis and tumor progression. However, the biological effects most circRNAs on cervical cancer remain unclear. Hsa_circ_0021087 (thereafter named circLMO1) is circRNA generated from circularization exon 2 3 LIM Domain Only 1 (LMO1) first identified as suppressor gastric cancer. Colorectal cancer (CRC) is the third most prevalent to be diagnosed, and it has a substantial mortality rate. Despite numerous studies being conducted on CRC, remains significant health concern. The disease-free survival rates notably decrease as CRC progresses, emphasizing urgency for effective diagnostic therapeutic approaches. development caused by environmental factors, which mostly lead disruption of signaling pathways. Among these pathways, Wingless/Integrated (Wnt) pathway, Phosphatidylinositol 3-kinase/protein kinase B/mammalian target rapamycin (PI3K/AKT/mTOR) Mitogen-Activated Protein Kinase (MAPK) Transforming Growth Factor-β (TGF-β) p53 pathway are considered important. These pathways also regulated non-coding RNAs (ncRNAs), including microRNAs (miRNAs), long (lncRNAs), circular (circRNAs). They have emerged crucial regulators gene expression in changing their levels. altered patterns ncRNAs been implicated progression development, suggesting potential targets. Ulcerative colitis (UC), an inflammatory bowel disease (IBD), may increase the risk of colorectal cancer (CRC) by activating chronic proinflammatory pathways. The goal this study was to find serum prediction biomarkers in UC CRC development combining low-density miRNA microarray and biocomputational approaches. expression profiles were compared microarray, finding five upregulated miRNAs specific progression (hsa-let-7d-5p, hsa-miR-16-5p, hsa-miR-145-5p, hsa-miR-223-5p, hsa-miR-331-3p). circRNA/miRNA/mRNA competitive endogenous RNA (ceRNA) network analysis showed that candidate connected well-known colitis-associated ACVR2A, SOCS1, IGF2BP1, FAM126A, CCDC85C mRNAs, circ-SHPRH circRNA. SST SCARA5 genes regulated hsa-let-7d-5p, hsa-miR-331-3p linked a poor survival prognosis patient dataset from Cancer Genome Atlas (TCGA). Lastly, our mRNA candidates validated comparing their differentially expressed mRNAs tissue databases. A high level parallel reduction SOCS1 found serum. We propose possibly hsa-let-7d-5p as novel for predicting CRC. More clinical sample is required further validation.

The stem-like properties cells were detected by evaluation stemness markers, sphere formation assay and flow cytometry. qRT-PCR, FISH, IHC, western blotting assessed expression localization circFNDC3B, RNF41, ASB6, markers CRC. metastatic capabilities examined wound healing Transwell assays, well vivo liver metastasis model. Bioinformatics analysis, RNA immunoprecipitation (RIP), pull-down co-IP used to detect associations among FXR2, ASB6. Downregulated was associated unfavorite survival patients, overexpression suppressed metastasis. Mechanistically, studies revealed that YTHDC1 facilitated cytoplasmic translocation m 6 A-modified enhanced RNF41 mRNA stability via binding FXR2. promoted ASB6 degradation through RNF41-mediated ubiquitination.
