Recently, biological aging has been quantified in DNA-methylation samples of older adults and applied as so-called "methylation profile scores" (MPSs) separate target samples, including children. This nascent research indicates that (1) can be early the life course, decades before onset aging-related disease, (2) is affected by common environmental predictors childhood development, (3) shows overlap with "developmental processes" (e.g., puberty). Because MPSs were computed using algorithms developed adults, these studies indicate a molecular link between environments, adult aging. Yet, if used to connect development aging, previous only traveled one way, deriving applying them Researchers have not yet epigenetic measures reflect pace child tested whether resulting are associated physical psychological In this perspective I posit combining new quantifications power address fundamental questions about span: How experience related adulthood? And what aging?

Abstract Background Epigenetic algorithms of aging, health, and cognition, based on DNA-methylation (DNAm) patterns, are prominent tools for measuring biological age have been linked to age-related diseases, cognitive decline, mortality. While most these methylation profile scores (MPSs) developed in blood tissue, there is growing interest using less invasive tissues like saliva. The aim the current study probe cross-tissue intraclass correlation coefficients (ICCs) MPSs applied saliva DNAm from same people. our primary focus that were previously found be robustly correlated with social determinants including second- third-generation clocks physiology we also report ICC values first-generation enable comparison across metrics. We pooled three publicly available datasets had both individuals (total n = 107, aged 5–74 years), corrected cell composition within each computed ICCs. Results after correcting composition, saliva–blood ICCs moderate indices aging cognition. Specifically, PCGrimAge highest (0.76), followed by PCPhenoAge (0.72), a measure performance (Epigenetic- g , 0.69), DunedinPACE (0.68), Acceleration (0.67), (0.66), an MPS hs-CRP (0.58), BMI (0.54). These appear lower than previous reports within-tissue (saliva range 0.67 0.85, 0.73 0.93). Cross-tissue acceleration measures poor, ranging 0.19 0.25. Conclusions Our findings suggest applying related phenotypes results similarity precise correspondence differs measure. degree several may suffice some research settings, it not suitable clinical or commercial applications. Abstract Background and Objectives Methylation profile scores (MPSs) index biological aging aging-related disease in adults are cross-sectionally associated with social determinants of health childhood. MPSs thus provide an opportunity to trace how biology responds environmental changes early life. Information regarding the stability life is currently lacking. Method We use longitudinal data from children adolescents ages 8-18 (N = 428, M age 12.15 years) Texas Twin Project. Participants contributed two waves salivary DNA-methylation (mean lag 3.94 years), which were used construct four reflecting multi-system physiological decline mortality risk (PhenoAgeAccel GrimAgeAccel), pace (DunedinPACE), cognitive function (Epigenetic- g ). Furthermore, we exploit variation among participants whether they exposed COVID-19 pandemic during course study participation, order test a historical period characterized by disruption might affect children’s MPSs. Results All showed moderate (test-retest r s 0.42, 0.44, 0.46, 0.51 for PhenoAgeAccel, GrimAgeAccel, Epigenetic- , DunedinPACE, respectively). No differences apparent between those whose second assessment took place after onset vs. whom both assessments prior pandemic.
