Obstructive sleep apnea (OSA) has been linked to disruptions in circadian rhythm and neurotrophin (NFT) signaling. This study explored the link between neuromodulators, chronotype, insomnia OSA. The participants (n = 166) underwent polysomnography (PSG) before being categorized into either control or OSA group. following questionnaires were completed: Insomnia Severity Index (ISI), Epworth Sleepiness Scale, Chronotype Questionnaire (morningness-eveningness (ME), subjective amplitude (AM). Blood samples collected post-PSG for protein level assessment using ELISA kits brain-derived neurotrophic factor (BDNF), proBDNF, glial-cell-line-derived factor, NFT3, NFT4. Gene expression was analyzed utilizing qRT-PCR. No significant differences found neuromodulator levels patients controls. controls with exhibited elevated gene (p < 0.05). In non-insomnia individuals, BDNF NTF3 increased group compared 0.007 both); there no groups. ISI scores positively correlated all expressions both groups, except NTF4 (R 0.127, p 0.172). AM ME predicting factors score clinically 0.05 groups). Compromised compensatory mechanisms may exacerbate insomnia. correlation chronotype NFT highlights role of misalignments disruptions.

Within the central nervous system, synaptic plasticity, fundamental to processes like learning and memory, is largely driven by activity-dependent changes in strength. This plasticity often manifests as long-term potentiation (LTP) depression (LTD), which are bidirectional modulations of efficacy. Strong epidemiological experimental evidence show that heart-brain axis could be severely compromised both neurological cardiovascular disorders. Particularly, disorders, such heart failure, hypertension, obesity, diabetes insulin resistance, arrhythmias, may lead cognitive impairment, a condition known cardiogenic dementia. Herein, we review available knowledge on molecular mechanisms dementia arise describe how LTP and/or LTD induction maintenance CA1 region hippocampus metabolic syndrome, arrhythmias. We also discuss emerging endothelial dysfunction contribute directly altering hippocampal impairing synaptically induced activation nitric oxide synthase. A better understanding CV disorders impact proper function synapses will shed novel light underpinnings dementia, thereby providing new perspective for more specific pharmacological treatments.

The brain-derived neurotrophic factor (BDNF) has become one of the cornerstones neuropathology, influencing synaptic plasticity, cognitive resilience, and neuronal survival. Apart from its molecular biology, BDNF is a powerful target for transformative benefit in precision medicine, leading to innovative therapeutic approaches neurodegenerative psychiatric diseases like Alzheimer's disease (AD), Parkinson's (PD), major depressive disorder (MDD), post-traumatic stress (PTSD). Nevertheless, clinical applicability obstructed by hurdles delivery, patient-specific diversity, pleiotropic signaling. Here, we summarize findings research, including regulatory pathways diagnostic/prognostic biomarkers integrative approaches. We describe delivery systems, such as lipid nanoparticle-based mRNA therapies CRISPR-dCas9-based epigenetic editing that bypass obstacles BBB (blood-brain barrier) enzymatic degradation. recent implementation multiplex panels combining biodynamic indicators with tau amyloid-β signaling markers showcases novel levels specificity both early detection potential monitoring. Humanized preclinical models iPSC-derived neurons organoids point key role neurodeveloping processes, paralleling advances bridging observation environments. Moreover, tools delivering TrkB activators or AI-based dynamic care platforms enable tailored scalable treatments. Neurotrophins (NTs) constitute a family of small protein messengers that play fundamental role in both the central and peripheral nervous systems. In particular, nerve growth factor (NGF) brain-derived neurotrophic (BDNF) subtle survival, differentiation, functioning neuronal populations, as well fine regulation immune functions. The SARS-CoV-2 infection was characterized by sequela symptoms (serious respiratory pathology, inflammatory storm, neurological discomfort, up to less serious flu-like symptoms), which caused, at end 2023, more than 7 million deaths worldwide. Despite official pandemic, physical psychological consequences are currently object scientific research, acute chronic/long-lasting (Long-COVID-19). Given multifactorial nature outcomes adults children, several studies have investigated potential involvement NGF BDNF systems pathology. This narrative review aims summarize most recent evidence on this crucial topic.
