Chloroquine (CQ) and its derivate hydroxychloroquine (HCQ), the compounds with recognized ability to suppress autophagy, have been tested in experimental works clinical trials as adjuvant therapy for treatment of cancers different origin increase efficacy cytotoxic agents. Such strategy can be effective overcome resistance standard chemotherapy or anti-angiogenic therapy. This review presents results combined application CQ/HCQ conventional drugs (doxorubicin, paclitaxel, platinum-based compounds, gemcitabine, tyrosine kinases PI3K/Akt/mTOR inhibitors, other agents) malignancies obtained experiments on cultured cancer cells animal xenograft models, a few examples trials. The effects such approach viability tumor growth, well autophagy-dependent independent molecular mechanisms underlying cellular responses are summarized. Although majority studies vitro vivo shown that effectively sensitize agents potential chemotherapy, often inconsistent. pharmacological suppression autophagy remains promising tool increasing development more specific is required.

The global increase in cancer cases and mortality has been associated with inflammatory processes, which chemokines play crucial roles. These molecules, a subfamily of cytokines, are essential for the migration, adhesion, interaction, positioning immune cells throughout body. Chemokines primarily originate response to pathogenic stimuli cytokines. They expressed by lymphocytes bloodstream divided into four classes (CC, CXC, XC, CX3C), playing multifaceted roles tumor environment (TME). In TME, regulate behavior recruiting such as tumor-associated macrophages (TAMs) myeloid-derived suppressor (MDSCs), promote survival. Additionally, they directly influence behavior, promoting pathological angiogenesis, invasion, metastasis. On other hand, can also induce antitumor responses mobilizing CD8+ T natural killer (NK) tumor, reducing pro-inflammatory enhancing responses. Given complex interaction between chemokines, system, angiogenic factors, metastasis, it becomes evident how important is target these pathways therapeutic interventions counteract progression. this context, physical exercise emerges promising strategy due its role modulating expression anti-inflammatory response. Aerobic resistance exercises have beneficial profile cancer, increased infiltration improvement intratumoral vasculature. This creates an less favorable growth supports circulation chemokines. Therefore, understanding impact on provide valuable insights treatment prevention.
