Clostridial dermatitis (CD), caused by Clostridium septicum , is an emerging disease of increasing economic importance in turkeys. Currently, there are no effective vaccines for CD control. Here, two non-toxic domains C . alpha toxin, namely ntATX-D1 and ntATX-D2, were identified, cloned, expressed Escherichia coli as recombinant subunit proteins to investigate their use potential vaccine candidates. Experimental groups consisted a Negative control (NCx) that did not receive challenge, while the adjuvant-only Positive (PCx), immunization (D1) ntATX-D2 (D2) received challenge. Turkeys immunized subcutaneously with 100 μg protein at 7, 8 9 weeks age along oil-in-water nano-emulsion adjuvant, followed challenge 11 age. Results showed 46.2% birds PCx group died post-challenge, rate mortality D1- or D2-immunization was 13.3%. The gross histopathological lesions skin, muscle spleen severity highest group, D2-immunized had significantly lower lesion scores when compared PCx. Gene expression analysis revealed higher pro-inflammatory cytokine genes than NCx D2 these Peripheral blood cellular increased frequencies activated CD4+ and/or CD8+ cells D1 groups. Additionally, turkeys developed antigen-specific serum IgY antibodies.
